摘 要:目的:探讨染色体着丝粒点(Cd)的变异与宫颈癌CasKi细胞和绒癌Bewo细胞染色体非整倍性畸变的关系。方法:肿瘤细胞株传代后用常规方法制备染色体,用直接法制备胚胎绒毛组织染色体标本,采用Cd-NOR同步银染分析技术研究CasKi细胞和Bewo细胞染色体Cd的变异。结果:与正常人胚胎绒毛细胞染色体Cd相比较,CasKi细胞染色体Cd缺失显著升高(P<0.0125)、Cd迟滞复制显著升高(P<0.0125)、Cd-NOR融合显著升高(P<0.0125),而小Cd没有显著性差异。与正常人胚胎绒毛细胞染色体Cd相比较,Bewo细胞染色体Cd缺失显著升高(P<0.0125)、Cd-NOR融合显著升高(P<0.0125),而Cd迟滞复制、小Cd没有显著性差异。结论:CasKi细胞染色体非整倍性畸变可能主要源于Cd缺失、Cd迟滞复制和Cd-NOR融合。Bewo细胞染色体非整倍性畸变可能主要源于Cd缺失和Cd-NOR融合。Objective:To study the relation between the centromeric dots(Cd)variation of CasKi cells and Bewo cells and the chromosome aneuploidy aberration.Methods: Chromosome of tumor cells were done with general method.Chromosome of normal embryonic villi were done straightly.A Cd-NOR in-phase argentum dye analysis technique was used to study centromeric dots(Cd) variation of CasKi cells and Bewo cells.Results: Frequency of Cd loss,frequency of Cd duplication laggard and frequency of Cd-NOR amalgamation in CasKi cells were significantly higher than those of normal embryonic villi(P0.0125).Frequency of little Cd showed no difference between CasKi cells and normal embryonic villi.Frequency of Cd loss and Frequency of Cd-NOR amalgamation in Bewo cells were significantly higher than those of normal embryonic villi(P0.0125).Frequency of Cd duplication laggard and frequency of little Cd showed no difference between Bewo cells and normal embryonic villi.Conclusion: Cd loss,Cd duplication laggard and Cd-NOR amalgamation might be related with aneuploidy aberration of CasKi cells.Cd loss and Cd-NOR amalgamation might be related with aneuploidy aberration of Bewo cells.
