摘 要:目的MicroRNAs(miRNAs)是一类在转录后调控基因表达的非编码RNA,越来越多的证据显示:一些在肿瘤中异常表达的miRNAs,有做为肿瘤诊断分子标记和判断预后的潜在价值。该研究应用miRNAs芯片技术筛选结直肠癌组织中差异表达的miRNAs,探索与结直肠癌相关的miRNAs。方法从3对手术切除的结直肠癌和相配对的正常结直肠组织中提取总RNA,miRNA芯片检测miRNA表达情况,然后在90例配对的结直肠癌和正常结直肠组织中,用Real-time RT—PCR(Taqman)对部分差异表达miRNA进行验证。结果与配对的正常结直肠组织相比,有22个miRNAs在结直肠癌中异常表达,其中miR-18a、miR-17和miR-20经Real—time RT-PCR(Taqman)验证,在结直肠癌组织中表达显著增高(P=0.006,0.023,0.034)。结论miR-18a、miR-17和miR-20可能参与了结直肠癌的发生过程。Objective MicroRNAs ( miRNAs ) constitute a class of small noncoding RNAs which controls gene expression at the posttranscription level. Emerging evidence suggests that miRNAs expression profiling in cancer may have potential value as prognostic and diagnostic biomarkers, but there is little data on global miRNA expression profiling in coloreetal cancer. The purpose of this study was to identify the miRNAs expression signature in human colorectal cancer. Methods Total RNA isolated from cancer tis- sues and corresponding noncancerous tissues from surgically resected colorectal cancers was extracted with Trizol reagent. The miRNA expression profiles in 3 pairs of colorectal cancer arid adjacent nontumorous tis- sue were analyzed using a mammalian miRNA microarray. The expression profiling of selected miRNAs was then investigated by Real-time RT-PCR ( Taqman ) in 90 pairs of colorectal cancer and adjacent nontu- morous tissue. Results Twenty-two miRNAs were identified by miRNA array analysis as being dysregu- lated in colorectal cancer tissues compared to normal tissues. Among them, the expression of miR-18 a, miR-17 and miR-20 were confirmed to be significantly higher in cancer tissues than in normal tissues (P=0.006, 0. 023, 0. 034). Conclusion In conclusion, the miR-18a, miR-17 and miR-20 overexpression may be involved in the development and progression of colorectal cancer.
