摘  要:目的建立测定山楂中绿原酸含量的高效液相色谱(HPLC)法。方法色谱柱为Sunfire C18柱(250 mm×4.6mm,5μm),流动相为甲醇-0.1 mol/L磷酸二氢钠缓冲液(25∶75),流速为1.0 ml/min,柱温为28℃,检测器波长为327 nm。结果绿原酸的质量浓度在7.03~70.3μg/ml范围内与峰面积值线性关系良好(r2=0.999 6),精密度RSD为0.27%,平均加样回收率为96.92%,RSD为0.92%(n=6)。结论 HPLC法简便易行、结果可靠,可以快速、准确地测定山楂中绿原酸的含量。Objective To establish a HPLC method for determination of chlorogenic acid in the hawthorn. Methods Sunfire CIS (4.6 mm × 250 mm,5 μm) column was used. The mobile phase was methanol: sodium dihydrogen phosphate(25 : 75). The flow rate was 1.0 ml/min, and the temperature was at 28℃. The detection wavelength was 327 nm. Results The calibration curve was at the range of 7.03 - 70.3μg/ml ( r = 0. 999 6), RSD was 0. 92% ( n = 6) and the average recovery was 96.92%. Conclusion The method was simple, reliable, accurate for determination of chlorogenic acid in the hawthorn.
