摘 要:目的 观察褪黑素对人肝癌 SMMC- 772 1细胞生长抑制作用及其作用机理。方法 采用不同浓度的褪黑素通过体外细胞培养、MTT显色技术研究其抑制作用 ,以流式细胞仪和电子显微镜技术观察其作用机理。结果 褪黑素对人肝癌 SMMC- 772 1细胞具有抑制作用 ,其 IC30 、IC50 分别为 1 .1 0和 2 .0 0 mmol/L。以 IC30 的褪黑素处理 SMMC- 772 1细胞 ,使其倍增时间由 2 1 .1 h延长到 38.4h。流式细胞仪观察到褪黑素导致 SMMC- 772 1细胞 G0 /G1期比例升高 ,S期比例降低。透射电镜可观察到以 IC30 的褪黑素处理的细胞 ,其超微结构发生了退行性改变。结论褪黑素对 SMMC- 772 1细胞具有抑制作用 ,其作用机理可能是褪黑素导致细胞 G0Objective This paper was to observe the inhibitory effect of melatonin on the growth of human liver carcinoma SMMC\|7721 line and its mechanism.Methods\ Inhibition of melatonin on the growth of human liver carcinoma SCCM\|7721 line was studied by examining the influence of melatonin soulution in warious concentrations on the cell culture in vitro with MTT assay.The mechanism investigation of the inhibitory effect was performed on flow cytometry(FCM) and transmission electron microscopy(TEM).JResults\ The 30% inhigitory concentration(IC 30 ) and 50% inhibitory concentration(IC 50 )were 1 10 and 2 00mmol/L respectively.In comparison with the case without treatment with melatonin the population doubling time was prolonged from 21 1h to 38 4h when the SMMC 7721 cells were treated with 1 10mmol/L melatonin.Examination with FCM found that the cell was inhibited by melatonin in G 0/G 1 phase and the cell percentage in S stage decreased.The ultrastructural features of cells treated by melatonin in 1.10mmol/L had degenerative changes when they were observed under TEM.Conclusion Melatonin could inhibit the growth of human liver carcinoma SMMC 7721 cells.The mechanism can be explained by the delay at the G 0/G 1 stage of the cell cycle caused by melatonin.
