血浆中miR-199a／b-3p表达水平在肝癌筛查及愈后诊断中的潜在价值

摘　　要：目的探讨血浆中miR-199a／b-3p表达水平是否可以应用于肝癌的筛查。方法采用real-timePCR方法及统计学方法分析肝癌患者（90例）和健康人（50例）的miR-199a／b-3p水平，分析肝癌手术前后（44例）miR-199a／b-3p表达水平，肝癌不同分期血浆中miR-199a／b-3p表达水平的差异，以及miR-199a／b-3p表达水平与肝癌患者存活期的相关性。结果肝癌患者血浆中miR-199a／b-3p表达水平明显低于健康人（t=-4．557，P〈0．01）；经手术切除肿瘤之后，患者血浆中miR-199a／b-3p表达水平较术前明显上升（t=-0．414，P〈0．01）；不同分期肝癌患者血浆中miR-199a／b-3p表达水平差异无统计学意义（F=0．258，P〉0．05）；术后miR-199a／b-3p表达水平与患者的存活时间正相关（r=0．788，P〈0．01）。结论通过检测血浆中miR-199a／b-3p表达水平可以对肝癌患者进行筛查，并对术后患者的愈后情况进行判断。Objective To evaluate the potential value of plasma level of miR-199a/b-3p in screening for liver tumor. Methods The differences of plasma level of miR-199a/b-3p between liver tumor patients（90 cases） and healthy controls （50 cases）, before and after surgeries（44 cases）, among different stages of liver tumor patients, were detected through real-time PCR statistical methods. The correlation of miR-199a/b-3p level and survival time of patients was also analyzed. Results The plasma level of miR-199a/b-3p was lower in liver tumor patients than in healthy controls （ t = - 4.557, P 〈 0.01）. The plaslna level of miR-199a/b-3p in patients after surgery to remove the tumor was significantly higher than before surgery （ t = - 0.414, P 〈 0.01）. The plasma levels of miR-199a/b-3p in 4 clinical stages were no statistical signifieanees（ F=0.258, P 〉 0.05）, and in postoperative patients and the survival time had a significantly positive correlation （ r = 0.788, P 〈 0.01 ） after operation. Conclusions The plasma level of miR-199a/b-3p can be used in screening liver tumor and predicting prognosis.
