摘  要:目的 探讨鼻咽癌HNE1细胞株端粒酶的表达。方法 采用TRAPPCRELISA法检测人HNE1、白血病HL6 0、肺癌TUL1、正常骨髓细胞 2例、培养的外周血单个核细胞及人血管平滑肌细胞株中端粒酶的表达。结果 人HNE1,HL6 0及TUL1细胞株中端粒酶均为阳性表达 ;其余 3种正常对照组细胞全部为阴性。HNE1细胞株热灭活后的端粒酶活性均为阴性 ,而在 10 2 个HNE1细胞中亦能检测到端粒酶活性。结论 端粒酶在鼻咽癌HNE1细株中具有高表达。端粒酶活性测定具有较高的特异性及灵敏度。Objective To study the expression of telomerase in the NPC HNE1 cell lines. Methods The telomerase activity was detected in HNE1 cell lines of NPC, HL60 cell lines of leukemia, TUL 1 cell lines of lung cancer, 2 cases of normal marrow cells, cultured mono-nucleus cells of peripheral blood, and vascular smooth muscle cell lines with TRAP PCR ELISA. Results High telomerase expression was detected in HNE1 cell lines, HL 60 and TUL 1 cell lines, but none in 3 kinds of normal control cells. After heat-inactivation, the telomerase activity of HNE1 cell lines disappeared obviously compared with that before heat-inactivation. By contrast, the telomerase activity in as few as 100 HNE1 cells could also be detecded. Conclusion There is a high telomerase activity in HNE1 cell lines and also a high sensitivity and specificity for telomerase assay.
