摘  要:对21种帘蛤科贝类线粒体16SrRNA基因片段的核苷酸序列进行了分析,以探讨这一序列在种质鉴定、分子系统发育研究中的应用价值。试验结果表明,所有物种扩增片段长度504~642bp,具有明显的长度多态性,序列A+T含量59.2%~69.0%,明显高于GC含量。物种间共有变异位点447个,其中简约信息位点351个。以16SrDNA片段序列为标记,以中国蛤蜊做外群,构建了帘蛤科贝类的系统发生树,拓扑结构显示,帘蛤科贝类形成3个明显类群,缀锦蛤亚科的13个种形成一个单系群,其结点自展值为93%。第二类群由雪蛤亚科、帘蛤亚科和镜蛤亚科的种类组成,结点自展值为87%。第三类群由仙女蛤亚科、青蛤亚科、楔形蛤亚科和文蛤亚科的种类组成,结点自展值为100%。结合拓扑结构分析和序列比对分析,本研究支持将短文蛤和丽文蛤订为文蛤的同物异名的观点,建议将丽文蛤和短文蛤订为文蛤的地理亚种;支持将薄片镜蛤和Dosinia angulosa订为2个独立种的观点;宜将波纹巴非蛤和织锦巴非蛤订为2个独立种。A nucleotide sequence analysis of mitochondrial 16S rRNA gene fragment was conducted among 21 clam species belong to family Veneridae to evaluate the utility of this sequence in identifying phylogeographic and phylogenetic structure.The length of 16S rDNA obtained was found to be 504~642 bp,with A+T contents from 59.2% to 69.0%,higher than G+C contents significantly.An analysis of sequence variation in the 16S rDNA revealed that there were 447 variable sites among 21 clam species in Veneridae,in which 351 were parsim-informative sites.The phylogenetic tree of Veneridae was reconstructed based on the nucleotide sequence of 16S rDNA,with Mactra chinensis as an outgroup species.The tree topology showed that the subfamily Tapetinae formed a monophyletic clade with bootstrap value(BPs) of 93%.The subfamily Chioninae,Venerinae and Dosininae formed another clade with BPs of 87%.The subfamily of Callistinae,Cyclininae,Sunettinae and Meretricinae formed the third clade with BPs of 100%.An analysis of tree topology and sequence alignment indicated that Meretrix lusoria,M.petechialis and M.meretrix were closely related species.The data support the viewpoint that M.petechialis and M.lusoria should be treated as synonym of M.meretrix and may be described as two geographical subspecies.By the data,Dosinia corrugata and D.angulosa are considered as two valid species,and it is validity that Paphia undulata and P.textile are two species.
