摘 要:目的肠源性内毒素血症是多器官功能衰竭的重要发病因素之一,而内毒素血症状态下肠蠕动变化及肠道神经内分泌系统的研究甚少。方法利用动物实验,观察内毒素血症状态下肠道传输速率变化:Wistar大鼠分为对照组、LPS组;观察内毒素血症后3、6、12小时大鼠血浆、空肠组织匀浆P物质含量变化及空肠免疫组化SP阳性神经在肠道分布及内毒素血症后的变化。结果内毒素血症后小肠传输速率加快;内毒素血症后血浆SP变化不显著,空肠SP含量下降,空肠SP阳性神经纤维及产物表达减少,且与空肠组织SP含量变化相一致。结论内毒素血症后肠道传输功能发生变化,可能与肠神经递质SP改变有一定关系。Objective: It has been known that enterogenic endotoxemia is an important pathogenetic factor in multiple or gan failure (MOF). However, the changes of intestinal transit and the neuropeptides in gut during endotoxemia is not well understood. Method: The effects of E. coli lipopolysaccharide (LPS) on the rate of small intestinal transit were investigated in Wistar rats which were given LPS 10mg/kg,intraperitonealy;The changes of neuropeptides during endotoxemia were investigated. The Wistar rats were divided into two groups: normal control and LPS group (LPS, 10mg/kg,intravenously). Tissue and serum levels of immunoreactive substance p (ISP) were measured at 3、 6、12 hours postendotoxemia. Immunohistochemistry methods were used to evaluate the effect of endotoxemia on the distribution of SP immunoreactivity of positive nerve fibre and product in the jejuna. Results: Intestinal transit was rapid during endotoxemia. LPS decreased the tissue levels of ISP without changes in peripheral plasma levels. Decreased SP immunoreactive in jejuna showed similar alterations in the tissue levels of neuropeptides. Conclusion: The changes of intestinal transit during endotoxemia possibly associated with SP.
