摘 要:目的评价TiC/Ti复合泡沫材料(TiC/Ti)、表面氧化处理的TiC/Ti复合泡沫材料(TiO2/TiC/Ti)及表面ZSM-5分子筛修饰的TiC/Ti复合泡沫材料(ZSM-5/TiC/Ti)对细胞增殖和分化的影响。方法在3种实验材料浸提液、阴性对照组泡沫碳化硅(SiC)浸提液和阳性对照组含0.64%苯酚的培养液中培养小鼠成纤维细胞L929,采用噻唑蓝(MTT)比色法比较材料浸提液中L929的细胞量。应用扫描电镜,以SiC为对照,观察小鼠成骨细胞MC3T3在3种实验材料上的生长状况,并在1、3、5 d分析比较各组材料上MC3T3细胞的碱性磷酸酶(ALP)含量。结果随着时间延长,3种材料浸提液中L929的细胞量均增殖,且L929细胞含量TiC/Ti>TiO2/TiC/T(i P<0.05)、TiO2/TiC/Ti>SiC(P<0.05)、SiC>ZSM-5/TiC/T(i P<0.05)。扫描电镜下,MC3T3细胞在3种实验材料表面均能贴壁黏附,形态正常,随着时间延长,数量增多,基质分泌增加。与对照组SiC比较,MC3T3细胞ALP的含量TiC/Ti>SiC(P<0.05)、SiC>ZSM-5/TiC/Ti(P<0.05)、TiO2/TiC/Ti稍高于SiC,但差异无统计学意义(P>0.05)。结论 3种实验材料均能促进细胞增殖和分化,与对照组SiC比较,促进细胞增殖的能力依次为TiO2/TiC/Ti>TiC/Ti>SiC>ZSM-5/TiC/Ti,而促进细胞分化的能力依次为TiC/Ti>TiO2/TiC/Ti>SiC>ZSM-5/TiC/Ti。Objective To study the effects of Surface morphology of TiC/Ti foam materials (TiC/Ti), Surface morphology of TiC/Ti foams with TiO2 eoatings(TiOJriC/Ti) and Surface morphology of TiC/Ti foams with molecular sieves(ZSM-5/TiC/Ti) on cell proliferation and dif- ferentiation. Methods MTF eolorimetric assay on L929 cell was carried out to compare the ceil counts in three groups with different ex- perimental materials leaching liquor with silicon carbide foam leaching liquor (negative control group ), and 0. 64% phenol solution ( positive control group). Take SiC as control, growth condition of MC3T3 ceil on the three materials was observed by scanning electron microscopy (SEM), and ceil alkaline phosphatase contents on the three materials were analyzed at day 1,3 and 5. Results L929 ceil counts in three materials leaching liquor were increased along with the time,and TiCfFi〉TiO2/TiC/Ti(P 〈 0.05),TiO2/TiC/Ti〉SiC(P 〈 0.05),SiC 〉ZSM-5/ TiC/Ti (P 〈 0.05 ). SEM showed that MC3T3 cells easily adhered to the three materials with normal morphology. Cell counts and the secre- tion of matrix were increased as time went on. Compared with SiC, MC3T3 cell alkaline phosphatase contents were significantly higher in TiC/ Ti (P 〈 0.05 ) and TiO2/TiCfFi (P 〉 0.05 ) groups, but lower in ZSM-5/TiC/Ti (P 〈 0.05 ). Conclusion All the three kinds of experi- mental materials can promote the proliferation and differentiation rate. Compare with SiC, the ability to promote cell proliferation is TiOJTiC/ Ti〉TiC/Ti〉SiC〉ZSM-5fFiC/Ti, and the ability to promote cell differentiation is TiC/Ti〉TiO2/TiC/Ti〉SiC〉ZSM-5/TiC/Ti.
