肺癌组织中bcl-2的表达及其与细胞凋亡和增殖的关系

摘　　要：目的　探讨凋亡相关基因bcl 2在肺癌中的表达及其与细胞凋亡和增殖的关系。方法　对 3 8例肺癌组织及 2 1例肺部良性组织 ,应用原位杂交技术检测了bcl 2mRNA表达情况 ;应用TUNEL技术对细胞凋亡情况进行了检测 ;应用免疫组织化学法对增殖细胞核抗原 (PCNA)等进行检测。并分别定义凋亡指数和增殖指数。结果　肺癌组织bcl 2基因及PCNA的表达明显高于肺部良性组织 ,P <0 .0 1;肺癌组织中凋亡指数和增殖指数 ( 3 .7%± 2 .4%,13 .9%± 15 .1%)分别高于肺部良性组织 ( 0 .8%±0 .5 %,0 .9%± 3 .2 %) ,P <0 .0 1;bcl 2阳性肺癌组织中凋亡指数 ( 1.9%± 1.2 %)明显低于bcl 2阴性肺癌组织 ( 5 .0 %± 2 .2 %) ,P<0 .0 1;它们的增殖指数均无显著性差异。结论　bcl 2在肺癌中的表达率较高 ,同时在肺癌中细胞凋亡和细胞增殖亦较活跃 ;bcl 2与细胞凋亡存在着密切关系 ,即bcl 2基因过度表达抑制细胞凋亡。bcl 2基因、细胞凋亡和PCNA均可作为肺癌的标志物。Objective To explore the expression of apoptosis related gene bcl 2 in lung cancer and the relationship of expression of bcl 2 to apoptosis and proliferation.Methods In 38 cancer cases and 21 benign respiratory cases,in situ hybrization technique was applied to detect bcl 2 mRNA, TUNEL technique was applied to detect the apoptosis and the immunohistochemistry was applied to detect PCNA.The apoptotic index(AI) and mitotic index(MI) was defined.Results The amount of bcl 2 mRNA of lung cancer tissues increased obviously, P <0.01.The AI and MI of lung cancer(3.7%±2.4%,13.9%±15.1%,respectively) were higher than those of the benign lung tissues(0.8%±0.5%,0.9±3.2%,respectively), P <0.01;The AI of cancer tissues with bcl 2 positive(1.9%±1.2%) was obviously lower than that with bcl 2 negative(5.0%±2.2%), P <0.01;Their MIs were not distinctly different.Conclusion The amount of bcl 2 mRNA increased obviously and the apoptosis and proliferation were active in the lung cancer,bcl 2 played an important role in bcl dependent apoptotic mechanism in lung cancer,the expression of bcl 2 can prevent apoptosis.bcl 2,apoptosis and PCNA can be used as the lung cancer molecular markers.
