上皮细胞激酶-2与层黏连蛋白5-γ-2在食管鳞癌细胞株TE13中相关性的研究

摘　　要：目的探讨上皮细胞激酶与层黏连蛋白laminin5-γ-2在食管鳞癌细胞株TE13中表达相关性。方法采用细胞免疫组化法检测EphA2与laminin5-γ-2在食管鳞状细胞癌细胞株TE13中的表达相关性;采用RT-PCR方法检测EphA2与LN5-γ-2mRNA表达的关系;采用Western Blot法检测食管鳞状细胞癌细胞株TE13中经RNA干扰技术特异性沉默EphA2后LN5-γ-2蛋白表达的变化。结果细胞免疫组化结果显示LN5-γ-2蛋白在细胞胞浆表达,且沉默EphA2后,LN5-γ-2表达减少。RT-PCR及Western Blot实验结果显示在食管鳞状细胞癌细胞株TE13中,LN5-γ-2mRNA及蛋白均随EphA2的沉默而受抑制(P<0.05)。结论 LN5-γ-2表达受EphA2的调控,可能为EphA2的靶基因。EphA2可能通过调控laminin5-γ-2对食管鳞癌肿瘤血管生成拟态进行调节。Objective To investigate the correlation of EphA2 and laminin 5-γ-2 in human esophageal cell TE13 line in vitro. Methods In the present study,human esophageal squamous cell carcinoma TE13 lines were examined for LN-5 expression immunohistochemically,using an antibody against LN-5-γ-2 chain.TE13 cell line with scilencing EphA2 gene were examined for LN-5-γ- 2 chain levels by Western blotting and RT-PCR. Results Laminin 5-γ-2 could be detected in cytoplasm in TE13 and the level of expression of lamanin 5-γ-2 decreased with silencing EphA2 immunohistochemically. RT-PCR and Western blot showed that the expression of laminin 5-γ-2 in TE13 was inhibited with silencing EphA2（P 0.05）. Conclusion The expression of laminin 5-γ-2 positively correlates with expression of EphA2.Laminin 5-γ-2 may be a potential target gene of EphA2.
