摘 要:目的通过对人垂体瘤转化基因1与存活剂在结肠癌组织中的表达的检测,探讨二者与结肠癌生物学行为之间的关系及其机制。方法回顾性分析了2007年12月~2011年12月笔者所在医院诊断为结肠癌的80例患者的临床资料,采用免疫组织化学法SP法对本组患者结肠癌组织标本中的hPTTG1和survivin蛋白的表达进行检测。结果 survivin在结肠癌组织中阳性表达率为82.5%(66/80),但是在正常的结肠黏膜组织中的阳性表达率为5.0%(1/20),差异有显著的统计学意义(P〈0.01)。hPTTG1在结肠癌组织中的阳性表达率为41.3%(33/80),但在正常的结肠黏膜组织中的阳性表达率为85.0%(17/20)。结肠癌组织中,hPTTG1与survivin的表达不存在显著的相关性(R=0.092,P〉0.05)。结论 survivin与hPTTG1在结肠癌组织中具有异常表达的现象,提示了二者在结肠癌的发生以及发展过程中所起的重要作用,可作为结肠癌发生及发展的一个"指示物"。Objective To detect the expression of human pituitary tumor transforming gene-1 and surviving expression in colon cancer tissue, and study the relationship with colon cancer. Methods 80 cases of patients diagnosed with colon cancer were retrospectively given analysis from December 2007 to December 2011, who stayed in the author's hospital, and used immunohistochemical staining to detect the expression of hPTTG1 and survivin of this group of colon cancer tissue specimens. And then it was analyzed that the relationship between hPTTG1 and survivin expression and colon cancer clinicopathological factors and prognosis. Results Survivin in colon cancer tissue positive expression rate was 82.5% (66/80), but the positive expression in normal colonic mucosa tissue was 5.0% (1/20), both existed statistically significant learning differences (P 0.01); the expression of survivin in colon cancer between gender, age, tumor location, tumor size, type, intestinal wall depth of invasion, histological grade, distant metastasis, Dukes' staging, etc. had no statistical difference (P 0.05). hPTTG1 in colon carcinoma, the positive expression rate was 41.3% (33/80), but it is the positive expression in normal colonic mucosa tissue was 85.0% (17/20), indicating that hPTTG1 in normal colon mucosal tissues, the positive expression rate was significantly higher than the rate of expression in colon cancer tissue, and had a statistically significant difference (P 0.01); the hPTTG1 expression and colon cancer tissue differentiation, Dukes stage and intestinal such as depth of invasion (P 0.05). Colon cancer tissue, hPTTG1 and survivin expression did not exist a significant correlation (R= 0.092, P 0.05). Conclusion Survivin in colon cancer tissue and hPTTG1 with abnormal expression of the phenomenon, which suggests the two played an important role in colon carcinogenesis and the development process. Therefore, both as colon cancer occurrence and development of an indicator.
