摘 要:目的探讨丹参酮ⅡA(TanⅡA)对脓毒症大鼠心肌炎性损伤的保护作用。方法采用盲肠结扎穿孔术制备脓毒症大鼠模型(CLP),然后再分别给予TanⅡA及生理盐水处理,于治疗12 h后观察血流动力学情况,检测血清cTnI及BNP、心肌细胞MPO、P-选择素基因表达、TNF-α和IL-1β水平。结果与假手术组相比,脓毒症大鼠血清中的cTnI及BNP、心肌细胞MPO、炎性因子水平和心肌P-选择素基因表达均明显升高(P均<0.05);TanⅡA治疗后可显著改善脓毒症大鼠的异常指标。结论 TanⅡA对脓毒症大鼠的心脏损伤具有较好的保护作用,可能通过降低心脏炎性水平和氧化应激反应来实现。Objective It is to explore the protecting effect of Tanshinone II A on the myocardial inflammatory injury in rats with sepsis. Methods The sepsis rat models were established by cecal ligation and puncture operation (CLP) , and the Tanshi- none Ⅱ A (Tan Ⅱ A) or normal saline was treated for 12 consecutive hours. Then, the following indexes were measured such as cTnI and BNP level in serum, myeloperoxidase, P - seleetin gene expression, TNF - α and IL - 1β level in myoeard. Results Compared with sham operated group, serum eTnI and BNP, myocardial inflammatory factor levels and myocardial myeloperoxidase activity, P - selectin gene expression all increased in the rats with sepsis ( P 〈 0.05 ) ; Tan lI A treatment could significantly improve the above aberrant indexes. Conclusion Tanshinone Ⅱ A shows a good protective effect on the cardiac in- jury in septic rats, which may be reached by improving inflammatory infiltration and oxidative stress reaction.
