摘 要:目的:改进大白兔VX2肝癌模型制作方法并研究肝癌组织缺血再灌注后的损伤作用。方法:用特殊技术方法制备高浓度细胞悬液,分别以27号注射针头向动物肝脏左中叶注射0.3 mL,制成动物肝癌模型。待肿瘤细胞种植成功至实验标准后,阻断肝左中叶之供血和门静脉60分钟,恢复血流至各个实验时间点。检测癌组织中CAT和SOD浓度的改变和细胞凋亡情况。结果:全部动物肝脏VX2细胞悬液注射后均种植成功但每组动物中有2-3只出现肿瘤细胞外溢和种植。种植成功之肿瘤组织缺血再灌注后CAT浓度明显降低且至1天达最低水平。SOD浓度在正常肝组织和肝癌组织中均明显降低而肝癌组织在再灌注后7天气浓度仍然保持较低水平。在正常肝组织和肝癌组织中肿瘤细胞凋亡均有增加,但在后者更为显著。结论:以特殊外科方法直接向肝组织内注射VX2细胞悬液能够成功制作大白兔肝癌模型。缺血再灌注可以导致组织的细胞凋亡,但对肝癌组织的影响更大。Objective: To improve the method of creating hepatocarcinoma models and evaluate injury of tumour tissues during is-chemia and reperfusion (IR). Methods: To achieve the tumour models, with special surgical technique, the viscous liver tumour cell sus- pension in a volume of 0.3 ml was injected into the left-middle lobe of liver parenchyma by using a 27-gauge needle during laparotomy. After tumour growth well to standard of experiment, the animals were subjected to 60 min clamp -induced ischemia of hepatic artery and portal vein in the left-middle lobe followed by reperfusion for different time. The changes of concentration of Catalase(CAT), super-oxide dismutase (SOD) and apoptosis in the tumour tissues were compared with that in the normal hepatic tissues. Results: All animals injected VX2 tumour cell suspension showed tumour growth and showed evidence of leakage and tumour seeding in 2-3 animals of each group. The concertration of CAT decreased markedly and reached the lowest level at reperfusion ld in hepatocarcinoma tissues. With SOD, two tissues types exhibited a significant decreasing in the concentration, but hepatocarcinoma tissues maintained this lower concentration for 7 d after reperfusion. The apoptotic cells increased in normal hepatic tissues and tumour tissue after IR, but the number of apoptotic cells in tumour tissue was lager than that in the normal hepatic tissues (P〈0.01). Conclusions: Direct injection of tumour cell suspension into liver parenchyma with special surgical technique may achieve good result of creating liver tumour moldes in the rabbits. IR induced the apoptosis of tumour tissue and normal hepatic tissues.
