摘 要:目的探讨人类白细胞抗原(human leukocyte antigen,HLA)-Ⅰ类基因A,B,C位点多态性与河南地区儿童急性淋巴细胞白血病发生的相关性。方法采用序列特异性聚合酶链式反应方法对河南地区98例急性淋巴细胞白血病患儿(观察组)和373例正常对照个体的(对照组)HLA-A,B,C位点的多态性进行检测。结果 2组受试者共检出HLA-A等位基因型16种,HLA-B等位基因型37种,HLA-C等位基因型15种;其中观察组检出HLA-A,B,C等位基因型分别13,26,13种,而对照组分别检出15,36,14种;HLA-B*07在观察组中的频率0.015明显低于对照组的频率0.056(P=0.026,OR=0.261,95%CI:0.080~0.850,χ2=8.734);除HLA-B*07外,HLA-A,B,C其他各等位基因在观察组和对照组中分布差异无统计学意义(P>0.05)。结论 HLA-B*07在河南儿童急性淋巴细胞白血病发生中可能起保护作用。Objective To explore the association of human leukocyte antigen (HLA) A, B and C polymorphisms with acute lymphoblastic leukemia in children in Henan province. Methods The polymorphisms of HLA-A, B and C were genotyped in 98 ALL patients and 373 healthy unrelated subjects by PCR-SSP method. Results Sixteen HLA A alleletypes, thirty-seven HLA-B alleletypes and fifteen HLA-C alleletypes were detected. The frequency of HLA B * 07 alleletype (0. 015) in AI.L patients was significantly lower than that in controls (0. 056) (P 0. 026, OR=0. 261, 950/00 CI.. 0.080 to 0. 850, X2 =8. 734). There were no significant differences in the other allele frequencies of HLA A, B and C between AI.L patients and controls (P〉0.05). Conclusion HLA-B * 07 may be a protective factor for the development of ALL in children in Henan.
