摘 要:肿瘤光动力治疗 (photodynamictherapy ,PDT)是现代癌症治疗中一种新的治疗法 ,为了更好地认识光动力诱导肿瘤细胞凋亡的作用机制。方法 :用Fluo 3AM荧光探针负载培养的人肺腺癌GLC 82细胞 ,在激光扫描共聚焦显微镜下观察细胞内Ca2 +浓度的变化。结果 :激光单独照射前后GLC 82细胞内Ca2 +的浓度变化不大 ;在PDT作用下 ,激光照射前GLC 82细胞内Ca2 +的浓度变化明显。结论 :本方法灵敏、简单、快速 ,能实时监测细胞内游离钙的变化。Objective:Photodynamic therapy (PDT) has become an important new modality in cancer treatment. In order to obtain a better understanding of the mechanisms of apoptpsis induced by PDT. Methods:Cultured GLC 82 cells were loaded with fluo 3/AM fluorescence probes, and the changes of intramyocytic Ca 2+ concentration were examined by laser scanning confocal microscopy. Results:there were negligible changes of intracytoplasmic calcium concentration of GLC 82 cells after laser irradiation alone. There were remarkble changes of intracytoplasmic calcium concentration of GLC 82 cells after laser irradiation. Conclusion: it is sensitive, rapid, simple and most effective method for real time quantitation of intracellular free calcium.
