摘  要:目的 进一步研究同型半胱氨酸(HCY)的致畸性及作用机制。方法 应用大鼠胚胎胚芽细胞微团培养法了解HCY对胚胎胚芽细胞增殖和分化的影响。结果 随着剂量的增加,HCY表现了对胚胎肢芽细胞的促进和抑制双相作用。半数抑制分化浓度(IC_(50))是5.0mmol/L,当加入S9时,IC_(50)减至3.5mmol/L;半数抑制增殖浓度是9.5mol/L,当加入S9时,IC_(50)为5.8mmol/L。结论 HCY对胚胎细胞分化的影响更为显著,对大鼠致畸作用不明显。Objective To determine potential mechanisms by which HCY may exerts its teratogenic effects. Method The micromass rat embryo limb bud cell culture was used to reveal the differentiation and proliferation of the cell under different HCY doses. Results With increased concentration, promotion effect changed into inhibition. The concentration inhibiting the formation of differential foci by 50% of the control is 5. 0 mmol/L and 3. 5 mmol/L when S9 was added; the concentration inhibiting the formation of proliferation by 50% of the control is 9.5mmol/L, when S9 is added , it was 5.8 mmol/L. Conclusion HCY obviously inhibited the cell differentiation than proliferation, while it is not a potent teratogen for rat.
