摘 要:探讨维甲酸对角膜基质创伤愈合的可能的作用。方法 :利用细胞计数法、 MTT法观察维甲酸对培养的人角膜基质细胞增殖的影响 ;利用缺损闭合实验观察维甲酸对培养的人角膜基质细胞移行的影响。结果 :MTT法和细胞计数法显示 ,维甲酸抑制体外培养的人角膜基质细胞增殖 ,抑制作用呈剂量、时间依赖性 ( F检验 ,P<0 .0 5) ,抑制率在 11.4 %~ 39.2 %。细胞计数法还显示 ,0 .5%苔芬兰染色示各组活细胞率均在 90 %以上 ,未见细胞毒性。缺损闭合实验表明 ,10 - 6 M维甲酸组缺损平均愈合天数明显长于对照组。结论 :维甲酸抑制培养的人角膜基质细胞增殖、移行。这些结论为进一步理解控制角膜基质细胞生长分化的机理 。Objective:To investigate the possible effects of retinoic acid (RA) on corneal stromal wound healing.Methods:By cell count and MTT techniques,the effects of RA on cultured human corneal keratocytes proliferation were evaluated;by measuring the defect close time,the influence of RA on cultured human corneal keratocytes migration was evaluated.Results:MTT and cell count showed that RA inhibited the proliferation of cultured human corneal keratocytes.The inhibition of proliferation was concentration dependent and time dependent.Inhibition rate was 11 4%~39 2%.Cellular viability was greater than 90% in all groups by trypan blue dye exclusion method.No cytotoxic effects were observed.The defect close time was signific -6 M RA group than that of control.Conclusion:RA inhibits the proliferation and migration of cultured human corneal keratocytes.These results lead to a better understanding of the mechanism that control differentiation and proliferation of corneal keratocytes,and provide a preliminary theoretical basis for the application of RA in controlling corneal wound healing and scar formation.
