摘　　要：目的探讨7-二氟甲氧基-5,4′-二-正辛烷氧基金雀异黄素(DFOG)对人乳腺癌细胞生长、凋亡的影响及其作用机制。方法体外培养人乳腺癌MCF-7和MDA-MB-453细胞。平皿集落形成法测定DFOG对乳腺癌细胞集落形成率的影响;碘化丙啶(PI)染色流式细胞术(FCM)检测细胞凋亡及分析细胞周期分布;Western blotting分析转录因子FoxM1及其下游蛋白CDK1、cyclin B、survivin、p27kip1表达。结果 DFOG以浓度依赖的方式抑制人乳腺癌MCF-7和MDA-MB-453细胞生长和诱导凋亡,并且伴随G2/M期细胞周期阻滞。DFOG能下调FoxM1及其下游蛋白CDK1、cyclin B、survivin表达,上调p27kip1蛋白表达。通过siRNA干扰技术下调FoxM1表达能增强DFOG对乳腺癌细胞的诱导凋亡作用。结论 DFOG显著抑制人乳腺癌MCF-7和MDA-MB-453细胞生长和诱导凋亡,下调FoxM1表达可能是其作用机制之一。ABSTRACT ： Objective genistein （DFOG） on the growth To investigate the effects of 7-difluoromethoxyl-5,4＇-di-n-octyl and apoptosis of breast cancer cells and its mechanisms. Methods Human breast cancer MCF-7 and MDA-MB-453 cells were cultured in vitro. The colony formation was detected by plate colony formation assay. The distribution of the phase in cell cycle and cell apoptotic rate were analyzed by using flow cytometry （FCM） analysis with PI staining. The expression of the transcription factor Forkhead Box M1 （ FoxM1 ）, and its downstream target proteins CDK1, cyclin B, survivin and p27Kip1 were detected by using Western blot. Results DFOG significantly induced the growth inhibition and cell apoptosis of MCF-7 and MDA-MB-453 cells in a concentration dependent manner. Further, DFOG efficaciously arrested the cell cycle in G：/M phase. DFOG decreased the expression of FoxM1 and its downstream genes, such as CDK1, cyclin B, survivin and increased p27kip1. The down-regulation of FoxM1 by siRNA before DFOG treatment resulted in enhanced cell apoptotic rate in MCF-7 cells.
