摘 要:目的探讨NOX1和NOX4在小鼠非酒精性脂肪性肝炎(NASH)发病中的作用。方法给予C57BL小鼠高脂饮食及脂多糖建立非酒精性脂肪肝模型,同时设立正常饮食组作为对照。观察各组小鼠血清ALT、AST值,HE染色评定脂肪性肝炎程度,实时荧光定量反转录-聚合酶链反应检测NOX1、NOX4mRNA的表达水平。结果非酒精性脂肪性肝炎模型建立成功,NASH模型组小鼠ALT、AST及肝指数明显高于高脂(HF)组(P<0.05),HF组NOX1、NOX4mRNA表达高于正常对照组(P<0.05),NASH组的NOX1、NOX4mRNA表达明显高于正常对照组和HF组(P<0.01)。结论本实验初步认为,NOX1和NOX4参与胰岛素抵抗的形成,可能是影响NASH发生和发展的重要因素。Objective To explore the role of NOX1 and NOX4 in mice with non - alcoholic steatohepatitis (NASH). Methods High fat diet and lipopolysaccharide were given to C57BL mice in order to establish NASH model, while the control group only received normal diet. Serum alanine and aspartate aminotransferase (ALT and AST) levels were observed. Real- time FQ -PCR method was used for testing of expression levels of NOXI and NOX4 mRNA. Results Model of NASH mouse was successfully established, ALT level, AST level and liver index in NASH group were higher than HF group (P 〈 0.05 ) , and expression of NOX 1 and NOX4 mRNA in HF group was higher than the control group(P 〈0.05) , furthermore among the 3 groups, expression of NOX1 and NOX4 mRNA in NASH group was the highest (P 〈 0.01 ). Conclusions The research proves that NOX1 and NOX4 partici- pate in the formation of insulin resistance, and this may be an important factor in the occurrence and development of NASH.
