摘　　要：目的检测新型转录抑制因子ZHX2基因mRNA在肝细胞癌(HCC)中的表达水平,分析探讨ZHX2mR-NA的表达及其与肝细胞癌发病机理之间的关系。方法建立了SYBR Green I实时荧光定量PCR方法,检测了正常肝组织、24例HCC及其癌旁组织中ZHX2基因的mRNA表达水平。结果 ZHX2基因的mRNA在6例正常肝组织中均有表达;在HCC癌组织中ZHX2mRNA的阳性率为33.3%,癌旁非瘤组织中ZHX2 mRNA的阳性率为62.5%;ZHX2mRNA在HCC组织中的表达明显低于对应的癌旁肝组织,差异具有显著性;将HCC按不同的临床病理特征分组比较ZHX2mRNA的表达,结果显示,ZHX2mRNA的表达与HCC患者的性别、年龄、分化程度、肿瘤大小、有无静脉浸润以及肝硬化程度无关,而与有无肝外转移灶、HBV感染和血清AFP值有关。结论 ZHX2mRNA表达量的减少与HCC的发生、发展以及侵袭转移密切相关。Objective To explore the relationship between transcriptional repressor ZHX2 expression and hepatocellular carcinoma genesis and development,the mRNA expression of ZHX2 in hepatocellular carcinoma（HCC） was studied by real-time PCR.Methods SYBR Green-based real-time PCR was established in order to assess the ZHX2 mRNA expression.And normal liver tissue and 24 cases of HCC and adjacent liver tissues samples were detected.Results The ZHX2 expression was detected in all 6 normal liver tissue samples,33% HCCs tissue samples and 62.5% adjacent tissues.The ZHX2 mRNA expression in HCCs tissue was less than adjacent tissues which showed significant difference.Grouping according to different clinical and pathological features,the ZHX2 expression was not related to gender,age,differentiation,tumor size,venous invasion and cirrhosis of the liver,but related to extrahepatic metastases,HBV infection and serum AFP value.Conclusion The reduced ZHX2 mRNA expression may play a role in Hepatocellular carcinoma genesis and development.
