摘 要:【目的】探讨沉默Pokemon基因对鼻咽癌细胞(CNE-2)放射敏感性的影响,寻求放射治疗鼻咽癌的新方法。【方法】首先利用siRNA预测软件得到3条siRNA序列,并制备合成siRNA片段,利用RT-PCR和免疫印迹法检测其沉默Pokemon基因的效果,筛选出沉默效果最佳的siRNA片段,转染CNE-2细胞后观察其对CNE-2细胞p53基因表达的影响、MTT法检测细胞的增殖活性、流式细胞术检测细胞的凋亡、克隆形成法检测转染siRNA联合放射处理后细胞的放射增敏比。【结果】成功筛选出了沉默效应最佳的siRNA片段,转染CNE-2细胞后能显著降低其Pokemon基因的表达、提高p53基因的表达、降低细胞的增殖活性、加速细胞的凋亡,同时增加了CNE-2细胞的放射敏感性。【结论】沉默Pokemon基因能增加鼻咽癌CNE-2细胞的放射敏感性,这为临床鼻咽癌的放射增敏治疗提供了新思路。[Objective ] To investigate the effect of pokemon gene knockdown on radiation sensitivity of nasopharyngeal carcinoma cell (CNE-2). [Methods] Three siRNA sequences were designed through siRNA prediction software, and the best siRNA was screened by detecting the expression of Pokemon with RT-PCR and Western blotting. After the siRNA was transfected to CNE-2 ceils, the p53 expression was investigated, and the cell proliferation was detected by MTF, and the apoptosis was detected by flow cytometry, and the radiosensitization ratio was detected by clone formation. [Results] The best siRNA was successfully screened. siRNA transfecting could decrease Pokemon expression, enhance p53 expression, reduce the cell proliferation, accelerate apoptosis, and augment the radiosensitization. [Conclusion] Knockdown of pokemon could increase radiosensitization of nasopharyngeal carcinoma cell (CNE-2), which supplied a new idea of the nasopharyngeal carcinoma radiotherapy.
