摘 要:于单因素试验基础上,以牡蛎蛋白和多糖提取率(Y1和Y2)为响应值,利用响应面法对牡蛎中蛋白和多糖提取工艺进行同步优化。试验结果表明,液料比(X1)、提取时间(X2)、提取温度(X3)及pH值(X4)4个因素对Y1和Y2均有显著影响。由响应面三维及等高线叠加图得到牡蛎蛋白和多糖提取率均高的最佳提取工艺参数:液料比33∶1,提取时间2.6h,提取温度40℃,pH值4.2。在此条件下,验证试验得到牡蛎蛋白提取率为21.15%,牡蛎多糖提取率为12.07%,与数学模型预测值非常接近。可见,响应面同步优化法对牡蛎蛋白和多糖提取条件进行同步优化合理可行。The extraction conditions of protein and polysaccharide from oyster Ostrea plicatula were optimized by response surface method with the protein and polysaccharide extraction yields (Y1 and Y2) as response values based on the single factor experiments. According to the models the all four factors including liquid to solid ratio, extraction time, extraction temperature and pH value were found to have significant influence on the protein and polysaceharide yields. The three dimensions and overlay contour plots revealed that the optimal conditions were found at concentration ratio of liquid to solid= 33 : 1, extraction time 2.6 h, extraction temperature 40 ℃, and pH value 4.2. Under the optimized conditions, the protein and polysaceharide extraction yields (Y1 : 21.15%; Y2:12.07 %) were found in close agreement with values predicted by the mathematics models, indicating that the response surface method is practicable for optimizing extraction process. This method improves the extraction conditions of protein and polysaccharide extraction from the oyster.
