摘 要:目的探讨不同剂量牛磺酸对锰染毒大鼠纹状体神经组织细胞损伤的影响。方法 SD健康雄性大鼠162只,随机分为对照组,10、15、20 mg/kg 3个剂量的染锰组和9个牛磺酸干预组,每组12只大鼠,余6只用于对照组、染锰组、牛磺酸干预组作电镜观察。干预12周后,开颅分离出大鼠纹状体,分别进行Tunel染色、透射电镜观察、多巴胺(DA)和5-羟色胺(5-HT)水平测定。结果染锰组和牛磺酸干预组纹状体细胞凋亡指数均高于对照组,差异有统计学意义(P<0.05);150、200 mg/kg牛磺酸干预对10、15、20 mg/kg染锰组大鼠纹状体细胞凋亡指数有降低作用,差异有统计学意义(P<0.05);大鼠纹状体透射电镜观察可见,15 mg/kg染锰组部分神经细胞出现核固缩等细胞凋亡的特征性改变,200 mg/kg牛磺酸干预可减轻其细胞凋亡程度;染锰组大鼠纹状体DA、5-HT水平均低于对照组(P<0.05),150、200mg/kg牛磺酸对10 mg/kg染锰组大鼠纹状体DA和5-HT水平增加效果显著(P<0.05)。结论锰可致大鼠纹状体神经组织细胞出现程度不同的细胞凋亡,降低纹状体中DA和5-HT的水平。牛磺酸在一定程度上对锰诱导的大鼠纹状体神经组织细胞凋亡及DA和5-HT的减少有保护作用。Objective To explore the effect of taurine on stfiatum tissue injury in manganese exposed rats. Methods 162 male SD rats were randomly divided into 1 control group, 3 manganese exposed groups ( 10, 15, and 20 mg/kg respectively) , and 9 intervened groups, with 12 rats in each group and 6 rats for electronic microscope examination. After 12 weeks of expo- sure, all rats were decapitated and their striatums were removed and observed by transmission electron microscope. DA and 5-HT levels were analyzed respectively. Results The striatum apoptosis indexes of the 3 dose groups exposed to manganese were sig- nificantly higher than those of the control group ( P 〈 0.05 ). Striatum apoptosis indexes of 9 intervened groups were significantly higher than those of the control group (P 〈 0. 05 ). 150 and 200 mg/kg of taurine could decrease apoptosis indexes of the groups exposed to 10, 15 and 20 mg/kg of manganeses respectively(P 〈0.05). The nucleus of striatum neurons in 15 mg/kg manga- nese exposed group became smaller, condensed, etc, which showed apoptosis of neurons. 200 mg/kg of taurine could decrease apoptosis of the group exposed to 15 mg/kg of manganese in ultrastructure. DA and 5-HT levels of 3 dose groups exposed to man- ganese were significantly lower than those of the control ( P 〈 0. 05 ). 150,200 mg/kg of taurine could significantly increase lev- els of DA and 5-HT in the group exposed to 10 mg/kg of manganese ( P 〈 0. 05 ). Conclusion Manganese could result in apop- totic morphology, increase level of apoptosis in striatum, and decrease levels of DA and 5-FIT. On a certain degree, taurine could protect neurons from apoptosis induced by manganese in vivo and improve the levels of DA and 5-HT.
