摘 要:为探明红托竹荪多糖的提取工艺及其体外抗氧化活性,采用水提醇沉法提取多糖,以多糖提取率为考察指标,通过单因素试验和正交试验,确定红托竹荪多糖的提取工艺条件;采用分光光度法测定多糖对羟自由基和超氧阴离子自由基的清除作用以及还原能力。结果表明,红托竹荪多糖提取的最佳工艺条件为料液比1∶25(g/mL),在80℃下提取3h,提取2次,红托竹荪多糖具有较好的还原能力,对.OH和O2-.具有较强的清除作用,其IC50值分别为0.51mg/mL和0.83mg/mL。结论:红托竹荪多糖具有明显的体外抗氧化活性。Abstract: To explore the extraction technology and in vitro antioxidation of polysaecharides from D. rubrovalvata. On the basis of single-factor experiments, the optimal extraction conditions of D. rubrovalvata polysaecharides by water extraction and alcohol precipitation were investigated with orthogonal experimental design. Meanwhile, the spectrophotometry was used to measure polysaccharides to hydroxyl free radicals and superoxide free radical scavenging effect and reducing power. The results showed that the optimal conditions were the ratio of solid-liquid 1 25 (g/mL), extraction temperature 80~C,extraction time 3 h, extraction twice. The reducing power was excellent and the D. rubrovalvata polysaccharides had powerful ability to clear off hydroxyl free radicals(ICs0 : 0.51 mg/mL) and superoxide free radicals(IC0:0.83 mg/mI.). Conclusion: D. rubrovalvata polysaccharides has significant in vitro antioxidation.
