抑癌基因p14^(ARF)对慢性粒细胞白血病细胞增殖、细胞周期及凋亡的影响机制研究

摘　　要：目的探讨抑癌基因p14ARF对慢性粒细胞白血病细胞的增殖、细胞周期及凋亡的的影响机制研究。方法应用慢病毒载体系统,将抑癌基因p14ARF导入慢性粒细胞白血病细胞系及患者来源的慢性粒细胞白血病细胞中,应用WST-8法检测细胞生长存活率,应用流式细胞术检测细胞周期和凋亡情况。结果应用表达p14ARF的VSV-G假构型慢病毒载体可明显抑制慢性粒细胞白血病细胞增殖,诱导细胞凋亡,但对细胞周期没有明显的影响。结论利用p14ARF基因治疗可明显抑制慢性粒细胞白血病细胞的增殖。Objective To explore the influencing mechanism of tumor suppressor gene p14ARF on the proliferation,cycle and apoptosis of chronic myeloid leukemia.Methods Used lentiviral vector transfer system to transduc tumor suppressor gene p14ARF into leukemia cell lines and leukemia cells sourced of patients,the survival rate of the cell was tested by WST-8 and the cell cycle and apoptosis by flow cytometry.Results VSV-G pseudotyped lentiviral vectors expressing p14ARF could inhibit leukemic cell proliferation apparently,and leaded to apoptosis.But p14ARF had no significant effect on the cell cycle in the chronic myeloid leukemia cells.Conclusion To use of p14ARF gene therapy can inhibit the proliferation of chronic myeloid leukemia cells apparently.
