摘  要:目的:建立SD大鼠正畸牙移动模型,观察牙移动过程中牙周组织内降钙素基因相关肽(calcitonin gene related peptide,CGRP)的表达变化,探讨其在正畸疼痛的外周神经机制中的作用。方法:将60只SD大鼠随机分为实验组(54只)和对照组(6只)2组,实验组大鼠施加50 g力后,分别在4 h、8 h、1 d(1 d组根据据力值大小分为1 d-30 g、1 d-50 g、1 d-80 g 3个亚组)、3 d、5 d、1周、2周,随机处死6只SD大鼠,收集牙周组织,进行实时定量PCR检测,观察牙周膜内CGRP随时间及力值大小的表达变化,采用SPSS13.0软件包对数据进行统计学分析。结果:大鼠正畸牙移动过程中,牙周组织内CGRP表达随加力时间发生改变,加力4 h后开始增强,1 d后达到高峰并逐渐减弱,2周后与对照组相比无显著差异。不同力值组加力1 d后,CGRP表达随加力力值增大而升高,80 g力组>50 g力组>30 g力组。结论:正畸牙移动过程中,牙周组织内CGRP表达随加力时间增加及加力力值增大出现规律性变化,提示CGRP可能在正畸疼痛的外周神经机制中具有重要作用。PURPOSE: To establish a Sprague-Dawley rat model of tooth movement, detect the expression of calcitonin gene-related peptide (ealeitonin gene related peptide, CGRP) in Sprague-Dawley rats' periodontal tissues during tooth movement and explore the role of the peripheral mechanisms of pain in orthodontics. METHODS: 60 Sprague-Dawley rats were randomly divided into experimental group (n=54) and control group (n=6). Force was applied on right maxillary first molar in rats and the changes of CGRP expression were detected by real-time PCR 4 h, 8 h, 1 d, 3 d, 5 d, 7 d, 14 d after tooth movement. SPSS 13.0 software package was selected and one-way ANOVA (least significant difference, LSD) was used for statistics analysis. RESULTS: Experimental tooth movement led to an increase in the expression of CGRP. In addition, following experimental tooth movement, the expression of CGRP in periodontal tissues was significantly up- regulated from 4 h, with a peak on 1 d and returned to the control level at 2-week. 1 d after experimental tooth movement, the application of different forces to rats induced a significant increase in CGRP expression. CONCLUSION: The regular change of CGRP expression in periodontal tissues indicates that CGRP may play an important role in peripheral mechanism of orthodontic pain.
