川芎嗪联合环胞霉素A逆转白血病多药耐药的研究

摘　　要：目的 :寻找有效的多药耐药联合逆转剂。方法 :用逆转录 -聚合酶链反应 (RT -PCR) ,Westernblot和MTT法研究川芎嗪逆转耐药细胞系HL - 6 0 /VCR的作用 ,对多种化疗药物耐药的白血病用川芎嗪与环孢菌素A(CsA)加化疗药物联合用于治疗 2 1例复发难治性白血病。结果 :单用川芎嗪在体外能不同程度地增强HL- 6 0 /VCR细胞对多种化疗药物的敏感性 ,降低p170蛋白的表达。川芎嗪联合环胞素A有较好逆转复发难治性白血病多药耐药的作用 ,治疗组完全缓解率 44 4% ,部分缓解 33 3%。结论 :川芎嗪联合环胞素A可能成为安全有效的逆转药物 ,在治疗复发难治性白血病中有较大的临床意义和推广价值。Objective: To search effective combination of multidrug resistance(MDR) modulators Methods:The reversal effects of tetramethylphylpyrazine(TMPZ)on the drug resistance of HL-60/VCR was evaluated by utilizing RT-PCR,Western blot and MTT Twenty one refractory and relapsed leukemia patients were received chemotherapy,combined with TMPZ and cyclosporin A(CsA) Results: Sensitivity of HL-60/VCR to multidrug chemotherapy was enhanced and p170 expression was decreased by the single application of TMPZ The reversal effectiveness of the TMPZ in combination with CsA to multidrug resistance of refractory and relapsed leukemia was favorable The complete remission(CR) rate was 44 4%and partial remission(PR) rate was 33 3%in the treatment group Conclusion: TMPZ in combination with CsA might be a safe and effective MDR reversing manipulation,and can be applied in the chemotherapy of refractory and relapsed acute leukemia
