食管癌多药耐药基因和多药耐药相关蛋白基因的研究

摘　　要：目的 :探讨食管癌组织中多药耐药基因 (mdr 1)和多药耐药相关蛋白基因 (mrp)表达的意义。方法 :运用逆转录 多聚酶链反应 (RT PCR)技术检测了 32例食管癌患者的癌组织标本及癌旁组织中mdr 1和mrp表达 ,分析其与肿瘤分化程度、浸润深度及淋巴结转移的关系。结果 :32例食管癌组织中mdr 1和mrp表达的阳性率均高于癌旁组织 (P <0 0 1,P <0 0 2 5 ) ,两基因的表达水平也显著高于癌旁组织 (P <0 0 0 1,P <0 0 0 1) ;在中低分化食管癌和肿瘤侵犯深肌层或纤维层时 ,mdr 1、mrp表达水平均高于高分化肿瘤和癌肿侵犯浅肌层以内者 (P <0 0 5 ,P <0 0 1)。结论 :食管癌组织中存在着原发性多药耐药性 ,mdr 1和mrp表达增高可能提示肿瘤的分化不良 ,并可反映肿瘤的浸润深度。Objective:To explore the expression of mdr 1 and mrp in tissue of esophagus cancer and analyse the relationship between their expression and tumor infiltration and lymphatic metastasis Methods:Using reverse transcription polymerase chain reaction (RT PCR) technique, we examined the positive expression of mdr 1 and mrp, and their levels of gene expression in 32 esophagus cancer samples Results:The positive rates of mdr 1 and mrp genes expression were 62 5%and 53 1%respectively in esophagus cancer and 28 1%and 25%respectively in the surrounded normal tissue The expression rates in cancer tissues were much higher than that in the surrounded normal tissue (P< 0 01,P< 0 025),and the levels of mdr 1 and mrp gene expression were also higher in cancer tissue than that in surrounded normal tissue (0 66∶0 27 and 0 46∶0 23, P< 0 001 and P< 0 001) The levels of mdr l and mrp expression in well differentiated cancer were lower than that in poorly differentiated cancer (P< 0 05) In cancer with deep muscle and fibro membrane infiltration,the levels of mdr l and mrp expression were also higher in comparison with invasion to shallow muscle Conclusion:Esophagus carcinomas express primary multidrug resistance genes High expression levels of mdr l and mrp might indicate undifferentiated tumor and reflect deeply infiltrating of the tumor
