SDF-1α促进BMSCs对脑缺血/再灌注大鼠CA1区神经元的保护作用

摘  要:目的观察基质衍生因子1α(SDF-1α)促进骨髓间充质干细胞(BMSCs)对脑缺血/再灌注大鼠CA1区神经元的保护作用。方法原代细胞培养BMSCs,传至第3代时以BrdU标记;以“四血管阻断”法制作大鼠脑缺血/再灌注模型,造模成功鼠随机分为模型对照组、BMSCs组和SDF-1α+BMSCs组。后2组经尾静脉分别注射BMSCs或SDF1α+BMSCs,模型对照组同法注射等量生理盐水,注射后分别存活1、3、7天和1个月,灌注固定制备石蜡切片,苏木精-伊红染色,光镜观察并计数各组大鼠海马CA1区锥体细胞在不同时间点的损伤表现。结果各组CA1区的形态异常细胞数高峰均出现于第3天,以后随时间延长而减少。但在各时间点内,BMSCs组和SDF-1α+BMSCs组CAl区形态异常细胞数均明显低于模型对照组,且SDF-1α+BMSCs组显著低于BM—SCs组,差异均有统计学意义(P〈0.05)。结论BMSCs移植可保护缺血/再灌注损伤大鼠海马CA1区的细胞损伤,且SDF-1α对这种保护有促进作用。Objective To investigate the protective effect of born nmrrow mesenchymal stem cells (BMSCs) promo- ted by stromal cell - derived factor -1α ( SDF -1α) to the nerve cells of hippocampal CA1 area cerebral ischemia/reper- fusion rats. Methods Cultured BMSCs and "four vessel occlusion" method were used for the cerebral I/R model. After reperfusion 24 h (hour), the saline (NS), BMSCs and SDF-1α+ BMSCs were respectively transplanted into the L/R model by the tail vein, and immunofluorescence determination was employed for examining the BMSCs of migrating to hip- pocampus at different time points of BMSCs transplantation. The models were divided into three groups: sham group, BMSCs group and SDF -1α + BMSCs group. The model animals were executed after transplanting 1, 3, 7 d and 1 M. Results The cell number of abnormal modality was a peak at 3 days after transplanting in each group , and it was de- creased following the time extended. In BMSCs group and SDF -1α + BMSCs group, the cell number of abnormal modal- ity were lower than the sham group at the same time after transplanting, and the SDF - 1α + BMSCs group was the lowest among three groups. The difference was statistically significant ( P 〈 0.05 ). Conclusion The transplantation of BMSCs can improve cellular damage of hippocampal CAI area after ischemia/reperfusion, while SDF -1α can promote the protection.
