摘  要:目的研究新型治疗性疫苗NTV的体内外宫颈癌杀伤作用。方法采用51Cr释放法研究脾淋巴细胞对宫颈癌细胞TC-1的体外杀伤活性,将正常小鼠体内激活的免疫活性细胞输给荷瘤小鼠,采用过继性细胞毒性T淋巴细胞(CTL)转移实验研究CTL的体内抗肿瘤作用。结果 CTL杀伤实验设置的效应细胞与靶细胞数量比分别为3:1、10:1及30:1,51Cr释放分析结果显示,疫苗NTV能够显著诱导脾淋巴细胞的肿瘤特异性杀伤作用,杀伤作用呈剂量依赖性变化,杀伤效率最高可达到53.5%;疫苗NTV在过继性CTL转移实验中的肿瘤抑制率为64.7%,上述结果均与阴性对照组有统计学差异(P<0.05)。结论疫苗NTV在体内外对宫颈癌细胞均具有良好的杀伤作用。Objective To evaluate cytotoxic effect of a novel therapeutic vaccine NTV on cervical cancer in vivo and in vitro. Methods 51 Cr release assay was used to study the cytotoxic activity of splenic lymphocytes targeted at TC-1 tumor cells.The method was used as a gold standard due to its good specificity and reproducibility.In addition,adoptive T cell transfer experiment was used to study the antitumor effect of cytotoxic T lymphocytes in vivo.Results In 51 Cr release assay,the ratio of effector cells to tumor cells(E∶ T) was 3∶ 1,10∶ 1 and 30∶ 1.The result showed that NTV could induce antitumor effect of splenocytes in vitro.Cytotoxic activity increased with the increase of E∶ T and dosage immunized with NTV,and the maximum cytotoxic activity was 53.5%.The result of adoptive T cell transfer experiment also showed splenocytes from mice immunized with NTV exhibited a good antitumor activity.The inhibition rate of NTV was 64.7% in adoptive T cell transfer experiment.The results mentioned above had a statistically significant difference compared with the control group(P 0.05).Conclusion The results show that the therapeutic vaccine NTV exhibits a high cytotoxic effect on cervical cancer in vivo and in vitro,and good prospects for the treatment of cervical cancer.
