摘 要:活性氧(reactive oxygen species,ROS)攻击蛋白质、脂类和DNA,对细胞造成直接损伤,其中对DNA的损伤作用包括链的断裂和碱基修饰,对鸟嘌呤的氧化作用产生8-羟基鸟嘌呤(8-hydroxygualline,oh8Gua)。人类8-羟基鸟嘌呤糖苷酶1(human 8-oxoguanine DNA glycosylase 1,hOGGl)是去除DNA中oh8Gua的重要酶。研究表明,hOGG1基因具有单核苷酸多态性的特征,基因突变可能影响hOGG1的酶活性,导致DNA修复缺陷。因此,hOGG1基因多态性可能影响机体发生肿瘤的危险性。Reactive oxygen species(ROS) can directly damage cells by oxidizing protein,lipids and DNA.ROS cause strands break and base modifications in DNA,including the oxidation of guanine residues to 8-hydroxygualline(oh8Gua).Human 8-oxoguanine glycosylase(hOGG1) is the key component responsible for the removal of oh8Gua in DNA.Previous studies have revealed the presence of several polymorphisms within the hOGG1 locus.Some Polymorphisms of hOGG1 gene have an effect on the activity of hOGG1 enzymes,then influence the capacity to repair the oxidative damaged DNA,and ultimately affecte the genetic toxicity and carcinogenesis induced by cancer-related substrates.Numerous studies reported that polymorphisms of hOGG1 gene were associated with the risk of several kinds of cancer.
