摘  要:为了探讨PDGF-D及其受体PDGFR-β在胃癌组织中的表达及其与血管生成的关系.采用免疫组织化学法检测88例胃癌手术切除标本中癌组织、癌旁组织及正常胃粘膜中PDGF-D/PDGFR-β的表达,采用CD34标记的微血管内皮细胞,计算微血管密度(MVD).分析PDGF-D/PDGFR-β的表达与胃癌临床病例特征及MVD的关系.研究发现胃癌组织中的PDGF-D/PDGFR-β的表达及MVD显著高于癌旁组织及正常胃粘膜(P<0.05).PDGF-D/PDGFR-β的表达及MVD的值与胃癌的TNM分期、浸润深度及局部淋巴结转移有关(P<0.05);MVD还与胃癌的直径有关(P<0.05),PDGF-D/PDGFR-β两者的表达呈正相关性(r=0.647,P<0.05).PDGF-D/PDGFR-β阳性组的MVD平均值高于PDGF-D/PDGFR-β阴性组(P<0.05).结果提示,PDGF-D及其受体PDGFR-β在胃癌组织中呈特异性的高表达,并与胃癌的MVD有关,可能参与胃癌的血管形成,在胃癌的进展过程中起着重要作用.To investigate the expressions of PDGF-D and its receptor PDGFR-β and their relationship with angiogensis in gastric carcinoma.Expressions of PDGF-D,PDGFR-β and CD34-stained microvessel density(MVD) were detected by immunohistochemical assay in gastric carcinoma tissues,adjacent tissues and nor-mal gastric mucosa from 61 cases.The correlations among PDGF-D and PDGFR-β expressions,MVD and clinic pathological features were analyzed.PDGF-D,PDGFR-β and CD34 in gastric carcinoma tissues were significantly higher than those in adjacent tissues and normal gastric mucosa(P0.05).They were related to TNM stage,histological differentiation,depth of inva-sion and lymphatic metastasis(P0.05),while MVD had relationship diameter of tumor(P0.05).A signifi-cant positive correlation between PDGF-D or PDGFR-β expressions and MVD was observed by Spearman correlation test(r=0.647,P0.05).The results suggested that PDGF-D and its receptor PDGFR-β are specifically over-expressed in gastric carcinoma and may be in-volved in the angiogenesis and play an important role in the initiation and progression in gastric carcinoma.
