摘　　要：以盘龙参为材料,采用CTAB法提取基因组DNA,探讨SRAP-PCR反应体系中模板DNA浓度对扩增的影响。结果表明:该CTAB法提取基因组DNA的OD260/OD280为1.81,浓度为4.35μg/μL。该CTAB法提取的基因组DNA纯度好,片段完整,无降解;在20μL反应体系中,模板DNA浓度为3.0 ng/μL时,重复性和稳定性较好,适于SRAP-PCR技术。Genomic DNA was isolated by CTAB method using Spiranthes sinensis as material,the influence of different DNA concentrations in SRAP reaction system on an amplification reaction was discussed.The result showed that the OD260/OD280 values of DNA obtained by the modified CTAB method was about 1.81,and concentration of DNA was 4.35 μg/μL.The genomic DNA in this experiment was purified and no significant degradation,in 20 μL reaction system,the suitable concentration of DNA was 3.0 ng/μL.
