酸性纤维素酶对壳聚糖的降解作用研究

摘  要:为确定特定酸性纤维素酶在降解壳聚糖制备壳寡糖的应用潜力,根据不同作用条件下壳聚糖的水解率,考察该纤维素酶降解壳聚糖的基本条件,具体结果为酶解温度50℃、pH 5.0、酶解时间4 h、纤维素酶用量为0.7 mL。在确定的最佳酶解条件下,考察了酶解过程中壳聚糖溶液粘度的变化,同时通过有机溶剂分级沉淀法,对纤维素酶降解壳聚糖的最终产物进行分析,发现最终产物中聚合度(DP)<8和8<DP<16的壳寡糖的含量分别为42.8%和42.9%。结果表明该纤维素酶具有制备壳寡糖的应用潜力。To identify the potential use of a certain acidi cellulase in chitooligosaccharide preparation, conditions for chitosan degradation for the acidic cellulase were investigated based on the extent of chitosan hydrolysis. Results indicated that the optimal temperature and pH was 50℃ and 5.0, respectively. And the enzyme concentration and reaction time was 0.7 mL and 4 h, respectively. Furthermore, the viscosity of chitosan solution was also investigated during the enzymatic hydrolyze process, and the main products was analyzed by using organic solvent precipitation method. Results indicated that chitooligosaccharides with a degree of polymerization (DP) of 8-16 and DP〈8 were the main products, they were 42.8% and 42.9% of the total products in weight, respectively. Based on these results, potential use of the acidic cellulase in preparation of chitooligosaccharides was confirmed.
