摘 要:目的观察丁苯酞(NBP)对血管性痴呆(VD)大鼠海马CA1区神经元长时程增强(LTP)的影响,并进一步探讨其分子机制。方法成年雄性SD大鼠30只,随机分成3组即对照组、VD组、VD+NBP组,每组10只。各组大鼠分别进行水迷宫实验(MWM)测试其空间认知功能。MWM后,分别记录麻醉状态下大鼠海马CA1区神经元LTP变化,比较3组大鼠海马神经元在高频刺激后的兴奋性突触后电位(EPSP)斜率变化;电生理记录后,大鼠断头取脑,制备脑冰冻切片,免疫组化测定海马CA1区NR2B受体的表达情况。结果 VD组大鼠有明显的空间认知障碍;丁苯酞治疗后,VD+NBP组大鼠的空间认知障碍得到显著改善。高频刺激后,VD组EPSP斜率增加百分比显著低于对照组大鼠(VD组vs对照组:13.9±1.6 vs 25.1±1.2,P<0.01);VD+NBP组EPSP斜率增加百分比显著高于VD组大鼠(VD+NBP组vs VD组:20.2±1.2 vs 13.9±1.6,P<0.05)。VD组NR2B受体的表达量显著低于对照组(P<0.01),VD+NBP组NR2B受体的表达量显著高于VD组(P<0.01)。结论丁苯酞可通过提高VD大鼠海马CA1区LTP而改善突触可塑性,从而改善空间认知障碍,其机制与丁苯酞上调VD大鼠海马CA1区NR2B受体的表达有关。Objective To investigate the effect of dl-3-n-Butylphthalide(NBP) on long-term potentiation(LTP) in CA1 hippocampal area of vascular dementia(VD) rat,and to explore the molecular mechanisms.Methods 30 male Sprague-Dawley rats were randomly divided into 3 groups,which were control group(n= 10),VD group(n= 10) and VD+NBP group(n= 10).The animal model of VD accepted the two-vessel occlusion(2VO) method.The behavior was measured using Morris water maze(MWM) test.The excitatory postsynaptic potentials(EPSP) in CA1 area was then recorded and the EPSP slope was used to measure LTP.After the electrophysiological experiment,all the rats were killed and their brains were isolated respectively.The expression of NR2B receptor in CA1 area of each rat was analyzed by immunohistochemical staining.Results VD model was successfully established in this study and the average percentage of EPSP slope increasing in VD group was decreased markedly compared with that in control group(VD group vs control group:13.9±1.6vs 25.1±1.2,P0.01).Except for improving the spatial learning and memory deficits of VD rats,NBP significantly enhanced EPSP slope in the hippocampus of VD rats,and the average percentage of EPSP slope increasing in VD+NBP group increased obviously(VD+NBP group vs VD group:20.2±1.2 vs 13.9±1.6,P0.05).NBP promoted the expression of NR2B receptor in the CA1 region of hippocampus of VD group(P0.01).Conclusion NBP can improve the LTP in CA1 hippocampal area of VD rats.The underlying mechanism is associated with its promotion effect on the expression of NR2B receptor in the CA1 region.
