摘 要:目的研究Ang-1、Ang-2在鼻咽癌组织中的表达情况与鼻咽癌淋巴管密度的相关性。方法用免疫组化法检测Ang-1、Ang-2在鼻咽癌组织及鼻咽部慢性炎症中的表达情况,用单克隆抗体D2-4 0标记鼻咽癌瘤内及瘤周微淋巴管。分析Ang-1、Ang-2与鼻咽癌临床病理参数与微淋巴管密度之间的关系。结果 Ang-1、Ang-2在鼻咽癌组织中阳性表达率分别为46.15%(24/52)、63.5%(33/5 2),明显高于鼻咽部炎性组织2 5.0%(3/1 2)、3 3.3%(4/1 2)(P<0.0 5)。Ang-1在鼻咽癌组织中的表达与性别、年龄、T分级及临床分期、有无淋巴结转移等因素均无明显差异;Ang-2在鼻咽癌组织中的表达与性别、年龄、T分级及临床分期等因素无明显差异,但在淋巴结转移组中的表达率83.9%(26/3 1)明显高于无淋巴结转移组3 3.3%(7/2 1)(P<0.0 1)。D2-4 0标记鼻咽癌微淋巴管阳性表达率达9 2.3 1%(4 8/5 2),并且在淋巴结转移组中瘤内及瘤周微淋巴管密度均高于无淋巴结转移组(P<0.0 5)。联合Ang-1、Ang-2在鼻咽癌组织中的表达与瘤内及瘤周微淋巴管密度均成正相关。结论Ang-2与鼻咽癌淋巴结转移密切相关,微淋巴管密度可作为判断淋巴结有无转移的重要指标之一。Objective To study the expressions of Ang-1 and Ang-2 in nasopharyngeal carcinoma (NPC) and their relationship with nasopharyngeal lymphatic vessel density. Methods Immunohistochemical method was used to detect the expressions of Ang-1 and Ang-2 in NPC and chronic inflammatory tissue of nasopharynx. The intratumoral and peritumoral micro-lymphatic vessels in NPC specimens were marked with monoclonal antibody D2- 40. And the relationship between the expressions of Ang-1 and Ang-2 with clinicopathologieal parameters and lymphatic microvessel density in NPC was analyzed. Results Tile positive expression rates of Ang-1 and Ang-2 in NPC were 46. 15% (24/52) and 63. 5% (33/52), which were significantly higher than those in the inflammatory tissues (25.0%, 33.3% ) ( P 〈 0.05 ). The expression of Ang-1 had no significant relationship with patients ' gender, age, and T stage, clinical staging and lymph node metastasis of the tumor. The expression of Ang-2 had neither significant relationship with gender, age, T stage and clinical staging, but its expression rate in the lymph node metastasis group (83.9% , 26/31 ) was significantly higher than that without lymph node metastasis(33.3% , 7/21 ) ( P 〈0. 001 ). D2-40 labeling displayed a positive expression rate of 92.31% (48/52) in lymphatic mierovessels of NPC. The intratumoral and peritumoral lymphatic vessel densities of the lymph node metastasis group were higher than those of the group without lymph node metastasis respectively ( P 〈 0. 05 ). Combined expressions of Ang-1 and Ang-2 were positively related with intratumoral and peritumoral lymphatic vessel densities in NPC. Conclusion Ang-2 is closely related with lymph node metastasis in NPC, and lymphatic vessel density is an important indicator to estimate lymph note metastasis of this tumor.
