摘 要:目的分析电化学测菌法用于牙周细菌定量检测的实用价值。方法选择牙龈卟啉单胞菌ATCC33277作为质控标准菌株,采集40份慢性牙周炎患者的龈沟液样本,分别运用电化学测菌法、细菌培养和实时定量PCR法对龈沟液样本中的牙龈卟啉单胞菌进行定量,比较3种定量方法的检测结果。结果细菌浓度在一定范围内,细菌浓度的对数与电化学测菌法的标准化阻抗值呈良好线性关系;电化学测菌法获得的细菌数多于细菌培养,少于实时定量PCR方法;电化学测菌法检测结果与细菌培养、定量PCR检测结果正相关(P<0.01)。结论电化学测菌法在一定细菌浓度范围内可用于牙周细菌定量检测。Objective To investigate the clinical value of the established electrochemical method for periodontal bacteria quantification. Methods Porphyromonas gingivalis ATCC33277 was selected as the reference strains. Then Porphyromonas gingivalis was detected in forty gingival crevicular fluid samples collected from ten patients with chronic periodontitis using the electrochemical method, culture method and real-time quantitative PCR. The results obtained from the three methods were compared. Results Within a certain concentration range, a good linear relationship was found between the normalized impedance change and the logarithmic value of the bacteria concentration. The electrochemical methods detected more number of Porphyromonas gingivalis than culture, but less than real-time quantitative PCR, and had positive correlation with culture and PCR method ( P 〈 0.01 ). Conclusion The electrochemical method could substitute conventional methods for periodontal bacteria quantification.
