杀鞘翅目幼虫Bt菌株YM-03的δ-内毒素基因定位

摘　　要：苏云金芽孢杆菌（Bacillusthuringiensis）的morrisoni亚种YM－03（8a8b）是本实验室分离的对鞘翅目幼虫有高毒力的菌株。经PCR产物分析它含有cry3A基因，产生68ku的毒素蛋白。经质粒图谱分析，该菌株含有3个质粒，其大小分别为83，72，9Mu。以cry3A基因的PCR产物片段为模板标记探针，与YM－03总DNA杂交，结果显示该菌的δ－内毒素基因定位于染色体上，有望构建出稳定遗传的广谱工程菌。Bt strain of YM-03 (8a8b) was isoated and characterized as Bacillus thuringiensis subsp. morrisoni.It is a high toxic to Coleoptera larva. Measurement by SDC-PAGE electrophoresis showed that the size of molecule of crystal protein is 68KD. Analysis of PCR products showed that it contains cry 3A gene. Analysis of total DNA by agarose electrophoresis showed that YM-03 strain contains three plasmids, i. e. 83, 72 and 93MD. One of the PCR preducts of the cry 3A gene was used as the template for synthesis radiolabeled probe. The probe was hybridized with the total DNA of YM-03. The results showed evidently that the crystal protein gene of YM-03 was located on the chromosomes.
