ER-α基因PvuⅡ多态性与子宫内膜癌ER阳性表达及血清E_2的关联性

摘　　要：目的:研究雌激素受体α(ER-α)基因PvuⅡ多态性与子宫内膜癌(EC)组织ER阳性表达、血清雌二醇(E2)水平3者间的相互关系。方法:选择60例EC患者为EC组和60例健康者为对照组,检测两组ER-α基因PvuⅡ多态性,检测EC组ER阳性表达及E2水平。结果:ER-α基因PvuⅡ多态性在EC组和对照组间差异有统计学意义(P<0.05),P等位基因患EC的风险是p等位基因的1.775倍(OR=1.775,95%CI:1.045～3.012);EC组PP基因型癌组织ER阳性表达明显少于Pp、pp基因型(P<0.05);在ER阳性表达的组织中,PP基因型ER阳性表达面积的百分比也明显<Pp、pp基因型(P<0.05);PP、Pp、pp 3种基因型的血清E2水平差异无统计学意义(P>0.05)。结论:ER-α基因PvuⅡ基因多态性与EC的发病有相关性,P等位基因可能是EC的遗传易感基因,PP基因型与癌组织ER阳性低表达具有关联性。Objective： To explore the correlative of Estrogen Receptor-ct （ER-a ） gene polymorphism and endometrial cancer （EC） ER positive expression, serum estradiol levels. Methods ： The polymorphisms of ER - a Pvu Ⅱ and endometrial cancer ER positive expression and serum estradiol were detected in 60 endometrial cancer patients and 60 healthy controls. Results ： There was significant differ- ence in frequencies of allele and genotype in ER - a Pvu Ⅱ polymorphism between EC group and control group （ P 〈 0. 05 ） . The relative risk suffered from EC of p allele was 1. 775 times of the P allele （ OR = 1. 775, 95% CI： 1. 045 ～ 3. 012） . The expression of ER - positive in PP genotype cancerous tissue of EC group was significantly less than Pp, and pp genotypes （ P 〈 0. 05 ） ; the percentage of the ER - posi- tive expression area in PP genotypes was also significantly less than that in Pp, pp genotype （P 〈 0. 05 ） ; the serum estradiol levels of PP, Pp, pp genotypes were no significant difference （ P 〉 0. 05 ） . Conclusion： ER - a Pvu Ⅱ polymorphism is associated with EC, and P allele may be a risk factor for EC; PP genotype and ER- positive cancer tissues with low expression has relevance.
