摘　　要：目的探讨RNA联合干扰(RNAico)对肝癌细胞株Hepa1-6中丝裂原调控子20(CDC20)和乙酰肝素酶(HPSE)表达的抑制作用。方法设计针对小鼠肝癌细胞Hepa1-6 CDC20和HPSE mRNA的小干扰RNA,以阳离子脂质体包埋后转染鼠肝癌细胞Hepa1-6,即RNAico;转染48 h后,采用RT-PCR和Re-al-Time PCR测定CDC20、HPSE mRNA表达的变化。结果联合干扰组CDC20和HPSE mRNA表达明显抑制,与其他各组相比,在统计学上有显著性差异(P<0.05)。结论 RNAico可同时显著抑制小鼠肝癌细胞Hepa1-6中CDC20和HPSE mRNA的表达。【Objective】 To evaluate the inhibitory effect of combined siRNA（RNAico） on the expression of CDC20 and heparanase（HPSE） mRNA of mouse Hepa1-6 hepatocarcinoma cell lines.【Methods】The siRNAs which aimed at CDC20 and HPSE mRNA of Hepa1-6,were designed and synthesized.Using cation lipofectamine 2000 embedding techniques,the siRNAs were transfect into Hepa1-6 cell lines for 48 hours,and then the expression of CDC20 and HPSE mRNA were detected by RT-PCR.【Results】Compared with normal and negative control groups,CDC20 and HPSE mRNA expression in the RNAico groups were obviously decreased（P〈0.05）.【Conclusion】RNAico has remarkable inhibitory effect on the expression of CDC20 and HPSE mRNA in mouse Hepa1-6 cell lines.
