摘 要:目的研究麦考酚酸(mycophenolic acid,MPA)对翼状胬肉成纤维细胞(pte-rygium fibroblasts,PFB)活性的影响,并对其相关机制进行探讨。方法体外培养PFB,MTT比色法、Brdu掺入法观察MPA对PFB增殖的抑制作用,Western blot检测PFB内核转录因子-κB(nuclear factor kappa B,NF-κB)P65及IκB-α蛋白的表达。结果 MTT比色法发现不同浓度的MPA和丝裂霉素C对PFB增殖均有抑制作用,差异均有统计学意义(均为P<0.05),且呈时间、剂量依赖性,半数抑制浓度下48h抑制率分别为(31.9185±1.2776)%、(31.1235±1.5826)%,72h抑制率分别为(50.2929±2.4574)%、(47.9729±0.6142)%。Brdu掺入法发现不同浓度MPA组积分光密度值与空白对照组比较,差异均有统计学意义(均为P<0.05),进一步证实MPA可抑制PFB的增殖;Western blot显示,MPA可抑制PFB胞核内P65表达(与空白对照组表达量1.5429±0.1427相比,MPA组表达量仅为0.8864±0.0728),并能增加胞浆IκB-α的表达(空白对照组为1.5592±0.2676,MPA组为2.1425±0.3052)。结论 MPA对PFB增殖有显著的抑制作用,其作用机制可能与其抑制PFB的NF-κB通路有关。ObJeetive To investigate the effects of mycophenolic acid (MPA) on pterygium fibroblasts (PFB) activity, and determine its relative mechanism. Methods PFB were cultured in vitro, the inhibitive effects of MPA on PFB proliferation were observed by lVlTr and Brdu fluorescence staining, and the expressions of nuclear factor- KB (NF-KB) P65 and IKB-a in PFB were examined by Western blot. Results MMT found that the different concentrations of MPA and mitomycin C had inhibitive effects on PFB proliferation with time- and dose-dependant manner, there were statistical differences ( all P 〈0.05) ,the inhibitive rates at 48 hours under ICS0 were ( 31. 918 5±1. 277 6)% and (31. 123 5± 1. 582 6)% ,at 72 hours were (50. 292 9± 2.457 4)% and (47.972 9±0. 614 2)%. Brdu fluorescence staining showed that there were statistical differences in integrated optical density in different concentrations of MPA groups com- pared with blank control group ( all P 〈 0.05 ), which further certified the inhibitory effect of MPA on PFB. Western blot showed that MPA significantly inhibited the expres- sion of P65 in nuclei of PFB ( the expression in MPA group was 0. 886 4±0. 072 8, and blank control group was 1. 542 9±0. 142 7)and increased the expression of IKB-a in cy- toplasm (the expression in MPA group was 2. 142 5±0. 305 2,and blank control group was 1. 559 2 s 0.267 6). Coneluslon MPA has inhibitive effects on PFB proliferation, and its mechanism may be related with PFB inhibitory effect on NF-KB pathway.
