摘 要:以6个牛品种(秦川牛、南阳牛、郏县红牛、鲁西牛、安格斯和夏南牛)共计717个个体为研究对象,采用PCR-SSCP方法与DNA测序相结合的方法,对PPARδ基因全部编码区及部分内含子区域的SNPs位点进行检测,并分析了SNPs位点与秦川牛的部分胴体、肉质性状的相关性。结果表明,在检测样本中,未检测到编码区的突变,仅在PPARδ基因第2内含子上发现了1个61 C→T的突变。χ2检验显示,在该位点,南阳牛和夏南牛处于Hardy-Weinberg平衡状态(P>0.05),其余4个群体均处于Hardy-Wein-berg不平衡状态(P<0.05);除夏南牛群体处于中度多态,其余5个群体均属于低度多态;相关分析表明,61 C→T突变与秦川牛的宰前活重和胴体重显著相关(P<0.05),KK基因型个体的宰前活重显著高于KL和LL基因型个体;KK基因型个体的胴体重也显著高于LL基因型个体。该位点突变可能是影响牛产肉性状的主效QTL或与之紧密连锁的辅助选择标记。The polymorphism of the entire coding region and partial intron region in PPARδ gene was detected by PCR-SSCP and DNA sequancing methods in a total of 717 individuals from six cattle breeds, namely, Qinehuan, Nanyang, Jiaxian Red, Luxi, Xianan and Angus, and its association with some production traits in Qinchuan cattle was also analyzed. The result only displayed a 61 C→T mutation in PPARB gone intron 2. This locus was at Hardy-Weinberg equilibrium (P 〉0. 05) in Nanyang and Xianan breeds except for other breeds. In the six breeds, Nanyang breed possessed moderate polymorphisms (0.25 〈 PIC 〈 0.50 ), while other populations possessed lower polymorphism ( PIC 〈 0.25 ). Correlation analysis showed that the locus had significant effects in slaughter weight and carcass weight ( P 〈 0. 05 ) in Qinchuan breed. Individuals with the genotype KK was significantly higher slaughter weight than those with gcnotypes KL and LL ( P 〈 0. 05 ). The individuals with the ganotype KK was much better than those with genotypc LL in carcass trait ( P 〈 0. 05 ). This locus might be a major QTN or tightly linked to that affect production traits, suggesting that it could be a candidate molecular marker for the beef cattle breeding.
