摘　　要：目的探讨DPC4蛋白在结肠癌组织中表达及其临床意义。方法采用免疫组化S-P法检测60例结肠癌标本中DPC4蛋白的表达,并探讨其与患者的生物学行为及预后的关系。同时选择30例癌旁正常结肠组织(距肿瘤边缘>5 cm)作为对照。结果 DPC4蛋白在结肠癌标本中的阳性率明显低于癌旁正常结肠组织(χ2=31.42,P<0.01)。结肠癌组织中DPC4蛋白表达水平随组织分化程度的增加而增加(P<0.01),随肿瘤大小、临床分期的提高和淋巴结的转移而呈逐渐下降趋势(P<0.01或P<0.05)。结论 DPC4基因在结肠癌的发生、发展、转移和预后中起重要作用,与患者的生物学行为和预后具有明显相关性,有望成为结肠癌基因诊断和治疗的新靶点。Objective To investigate the expression of DPC4 protein in colon cancer and its clinical significance. Methods The expression of DPC4 protein was detected by immunohistochemical method in 60 cases of colon cancer, and its relationship with biological behavior and prognosis. Another 30 specimens of adjacent normal colon tissue （from the edge of the tumor〉5 cm） were taken as the control group. Results The expression of DPC4 protein in colon cancer tissues was significantly lower than that in adjacent normal tissue （X2=31.42,P 〈 0.01）. It showed a gradually increasing trend with the degree of differentiation in colon cancer （P 〈 0.01）. But it showed a gradually decreasing trend with clin ical degree, tumor size and lymph node metastasis in colon cancer （P 〈 0.01 or P 〈 0.05）. Conclusion DPC4 gene par ticipates in the development, progression, metastasis and prognosis of colon cancer, and has an obvious relation with biological behavior and prognosis. DPC4 gene may become a new target for anticancer gene diagnosis and therapy.
