摘　　要：建立了免疫亲和柱层析净化-高效液相色谱法(HPLC)检测小麦中DON的分析方法。小麦试样用水萃取,提取液稀释过滤后经过含有呕吐毒素特异性杭体的DON Test-P免疫亲和柱层析净化,甲醇洗脱,洗脱液氮吹至干,用流动相乙腈-水定容,用Cl8色谱柱分离,采用紫外检测。结果表明,当呕吐毒素质量浓度为0.1～5.0mg/L时,色谱峰面积与呕吐毒素质量浓度呈线性关系,方法的检出限为0.02mg/L,试样的平均回收率为84.0%～92.0%,测定值的相对标准偏差均小于5.0%。DON in wheat was determined by HPLC with immunoaffinity chromatography purification. The wheat samples were extracted with water. The filtrate was purified by passing through the DON Test-P immunoaffinity column, methanol elution, nitrogen blowing to dry and constant volume with the mobile phase of acetonitrile-water, using C18 column for separation and UV detection. The results showed that： linear relationship between values of peak area and mass concentration of DON was obtained in the range of 0.1-5.0 mg/L. Detection limit was 0.02 mg/L. Tests for average recovery rate were in the range of 84.0% -92.0%,and the relative standard deviation of the measured values were less than 5.0%.
