摘 要:目的:观察牙周组织形成过程中上皮剩余细胞的组织形态、细胞增殖凋亡。方法:TUNEL(TdT-mediated-dUTPnickendlabeling,TUNEL)法原位细胞凋亡检测牙周组织发育中上皮剩余细胞凋亡的情况。SP免疫组化法检测增殖核抗原(PCNA)和凋亡抑制基因Bcl-2、骨桥素(OPN)和骨涎蛋白(BSP)的表达。结果:牙周膜牙骨质侧可见上皮剩余(theepithelialrestofmalassez)沿牙骨质表面成簇分布,根分叉和牙颈部较多,细胞数量不等,可见不规则核,簇内细胞相互重叠、排列无规则,TUNEL结果示上皮剩余中可找到凋亡阳性表达细胞。免疫组化可见上皮剩余细胞阳性表达OPN,但PCNA和BSP在此期上皮剩余细胞中为阴性表达。结论:在牙周发育过程中,伴随牙槽骨的改建和牙周膜纤维束走向的调整,上皮剩余经历塑形过程,细胞凋亡在这一过程中发挥重要作用。上皮剩余细胞的OPN阳性表达提示其在此过程中可能参与牙骨质的发育。Objective:To observe the morphology,apoptosis,proliferation and distribution of epithelial rests of Malassez(ERM)and expression of bone-related noncollagenous glycoproteins in ERM,so as to evaluate characters and roles of ERM in the developing periodontium.Method:Apoptosis was identified by the terminal deoxy-transferase (TdT)-mediated dUTP-biotin nick end labeling (TUNEL) method.SP immunohistochemical method was used to detect the expression of bcl-2 protein,PCNA (proliferating cell nuclear antigen),OPN(osteopontin) and BSP(bone sialoprotein) in ERM.Result:ERM were observed as clusters of cells,mostly located in the cervical and furcational regions in the periodontal ligament of mouse molars,next to the cementum surface.The number of cells is uncertain,irregularly shaped nuclei could be found in ERM.Some cells of ERM contained dense round/ovoid structures,strongly stained by hematoxylin.Positive cells of TUNEL and OPN could be found in ERM,but the expression of PCNA and BSP in ERM were negative.Conclusion:The epithelial cells of Malassez’s rests undergo turnover or remodeling,along with reconstruction of alveolar bone and periodontal membrane fiber in the developing periodontium,apoptosis and OPN play important roles during the progress.
