四味肝泰软胶囊对人肝癌细胞株SMMC-7721细胞周期及凋亡影响的初步研究

摘　　要：目的:初步探讨不同浓度四味肝泰软胶囊对人肝癌细胞株SMMC-7721细胞周期及凋亡的影响。方法:采用流式细胞仪PI染色法和Annexin V-EGFP/PI双染法研究不同浓度四味肝泰软胶囊作用于人肝癌细胞株SMMC-7721后细胞周期DNA含量变化及对细胞凋亡率的影响。结果:各浓度四味肝泰软胶囊均可以调节G1/S转换,使肿瘤细胞发生S期阻滞,造成S期细胞堆积,阻断细胞的DNA合成和复制,阻滞肿瘤细胞进入G2/M期,从而起到抑制肿瘤细胞的增殖,诱导SMMC-7721细胞凋亡,且这种诱导效应具有明显的时间-剂量-效应关系。结论:四味肝泰软胶囊对人肝癌细胞株SMMC-7721具有明显的抑制作用,其作用机制可能与抑制肿瘤细胞DNA的合成和诱导细胞凋亡有关。Objective：To study the cell cycle and apoptosis of human hepatoma cell line SMMC 7721 induced by different concentra tions of Si-Wei Garl Tai soft capsules. Methods ： Flow cytolnetry PI staining and Annexin V-EGFP/PI were used to assay the DNA concentration and the apoptosis rate in hepatoma cell line SMMC 7721 after the Si-Wei Gan Tai of different concentration were used. Results： Si-Wei-Gan-Tai can regulate the G1/S transition of the hepatoma cell, which could make the cell in S phase arrest, resul ring in the accumulation of cells in S phase, blocking DNA synthesis and replication of cells, block tumor cells into the G2/M phase restrain the proliferation of tumor cells, and induced SMMC-7721 cells apoptosis, and then obvious relationship in time dose effect was confirmed by the inductive effect. Conclusion：Si Wei Gan Tai could inhibit the hepatoma cell line SMMC-7721 in vitro by in hibiting cell proliferation and inducing apoptosis of the cell.
