摘 要:目的探讨依维莫司(RAD001)联合顺铂对人胃癌SGC7901细胞凋亡及Bcl-2、Bax表达的影响。方法体外培养SGC7901细胞,将其分为对照组,顺铂组(顺铂2.50 mg/L),RAD001低浓度组(RAD001 5.00nmol/L)、RAD001中浓度组(RAD001 10.00nmol/L)、RAD001高浓度组(RAD001 20.00nmol/L)及联合组(顺铂1.25mg/L+RAD001 5.00nmol/L),采用流式细胞术检测SGC7901细胞的凋亡率,采用免疫细胞化学、Western blot检测SGC7901细胞Bcl-2和Bax的表达情况。结果 RAD001、顺铂作用SGC7901细胞48h后,对照组,顺铂组,RAD001低、中、高浓度组及联合组的细胞凋亡率分别为(8.04±0.48)%、(18.94±0.75)%、(10.47±1.05)%、(13.93±2.45)%、(17.20±0.65)%及(23.18±1.05)%。除RAD001低浓度组细胞凋亡率与对照组比较,差异无统计学意义(P>0.05),其他各组细胞凋亡率均较对照组显著增加(P<0.05)。与对照组比较,顺铂组、RAD001组及联合组SGC7901细胞Bcl-2蛋白表达下降,而Bax蛋白表达增加,Bcl-2/Bax比值降低(P<0.05),联合组改变最明显。结论 RAD001联合顺铂可通过上调Bax和下调Bcl-2蛋白的表达诱导SGC7901细胞凋亡。Abstract:Objective To investigate everolimus(RAD001) combined with cisplatin on apoptosis and Bcl-2, Bax expression of human gastric cancer SGC7901 cells. Methods SGC7901 cells were cultured in vitro and divided into control group,cisplatin group (cisplatin 2.50 mg/L),RAD001 low concentration group(RAD001 5.00 nmol/L),RAD001 medium concentration group(RAD001 10.00 nmol/L), RAD001 high concentration group (RAD001 20. 00 nmol/L) and combination group (cisplatin 1. 25 mg/L-+ RAD001 5.00 nmol/L). Flow cytometry was employed to detect apoptotic rate of SGC7901 cells and immunocytochemistry and Western blot were conducted to measure Bcl-2,Bax expression of SGC7901 cells. Results After 48 h treatment,apoptotic rates of SGC7901 cells in control group, cisplatin group, RADO01 low concentration group, RAD001 medium concentration group, RAD001 high concentration group and combination group were (8.04 ± 0.48 ) %, ( 18.94 ± 0.75) %, ( 10.47 ± 1.05 ) %, ( 13.93 ± 2.45 ) %, (17.20± 0.65) % and (23. 18± 1.05)%. Except RAD001 low concentration group showing no statistically significant difference when compared to control group, the apoptotic rates in the other groups were obviously increased(P〈0.05). Compared with con- trol group,SGC7901 cells in cisplatin group,RAD001 groups and combination group showed decreasing Bcl-2 and increasing Bax protein expression,and lower Bcl-2/Bax ratio(P〈0.05). Changes in combination group were the most obvious. Conclusion RAD001 combined with cisplatin can induce apoptosis in SGC7901 ceils via up-regulating Pax and down-regulating Bcl-2 protein expression.
