摘 要:目的观察银杏叶提取物(GBE)对硫代乙酰胺(TAA)所致肝损伤大鼠的肝脏保护作用及其对肝组织Caspase-3、Caspase-8表达的影响。方法将70只Wistar大鼠随机分为正常对照组(n=10)、模型组(n=15)及GBE组(n=45),GBE组再分为GBE低剂量组(n=15)、GBE中剂量组(n=15)及GBE高剂量组(n=15)。GBE低、中、高剂量组大鼠分别灌服GBE 50、100、200mg/kg,1次/d,连续30d;正常对照组及模型组大鼠灌服生理盐水。模型组及GBE低、中、高剂量组大鼠于灌服第28天用TAA(300mg/kg)行腹腔注射,隔天注射1次,共3次;正常对照组大鼠腹腔注射生理盐水。所有大鼠均于末次注射后24、48、72h采血,检测血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)水平。用逆转录PCR检测大鼠肝组织Caspase-3、Caspase-8mRNA的表达情况,并对Caspase-3、Caspase-8的活性进行检测。结果与正常对照组比较,模型组大鼠血清ALT、AST水平明显升高,肝组织Caspase-3、Caspase-8mRNA水平上调;而与模型组比较,GBE组大鼠血清ALT、AST水平明显降低,肝组织Caspase-3、Caspase-8mRNA水平下调。结论 GBE可通过调节Caspase-3、Caspase-8的表达抑制急性肝功能衰竭患者的肝细胞凋亡,从而对肝损伤发挥保护作用。Objective To observe the protective effect of ginkgo biloba extract(GBE) on thioacetamide(TAA)-induced hepatic injury in rats and its impact on the expression of Caspase-3 and Caspase-8 in hepatic tissue. Methods Seventy Wistar rats were ran- dornly divided into normal control group(n= 10) ,model group(n= 15) and GBE group(n=45) ,and the GBE group was subdivided into GBE low-dose group ( n = 15 ), GBE medium-dose group ( n= 15 ) and GBE high-dose group ( n = 15 ). Rats in the GBE low-, medium- and high-dose group were fed with GBE 50,100,200 mg/kg,respectively,once a day for 30 consecutive days,while rats in the normal control group and model group were fed with physiological saline. On the 28th day of feeding,rats in model group and GBE low-,medium-,high dose group were intraperitoneally injected with TAA(300 mg/kg) every other day with total of 3 times, while rats in normal control group with physiological saline. Blood collection was conducted in all rats at 24,48 and 72 h after last injection, and levels of serum alanine aminotransferase (ALT) and aspartate aminotransferase(AST) were detected. Reverse transcriptase-PCR was utilized to assay the expression of Caspase-3, Caspase-8 mRNA in hepatic tissue of rats and activities of Caspase- 3,Caspase-8 were measured. Results Compared to normal control group, levels of serum ALT, AST and Caspase-3, Caspase 8 mRNAexpression in hepatic tissue of rats in model group were significantly increased, however, when compared to model group, those in GBE group were significantly decreased. Conclusion GBE can inhibit apoptosis of hepatocytes in patients with acute liver failure via regulation of Caspase-3,Caspase-8 expression and play a protective role in hepatic injury. K
