S6K1和4EBP-1蛋白在膀胱尿路上皮癌中的表达及意义

摘　　要：目的探讨S6K1和4EBP-1蛋白在膀胱尿路上皮癌中的表达及意义。方法使用免疫印迹法(Western-blot)检测S6K1和4EBP-1蛋白在80例膀胱尿路上皮癌及20例正常膀胱黏膜中的表达。结果膀胱尿路上皮癌中S6K1蛋白表达率(64%)高于正常膀胱黏膜(10%),4EBP-1蛋白表达率(71%)高于正常膀胱黏膜(15%)。S6K1和4EBP-1蛋白在低分级膀胱癌中的表达率显著高于高分级膀胱癌中的表达率。复发肿瘤的S6K1和4EBP-1蛋白表达率高于初发肿瘤。而S6K1和4EBP-1蛋白表达率在不同性别患者、不同肿瘤分期和是否吸烟患者中差异无统计学意义。结论膀胱尿路上皮癌中S6K1和4EBP-1蛋白与膀胱癌的分级和复发相关,可能成为肿瘤诊断治疗的新靶点。Objective To detect the expression of S6K1 and 4EBP-1 and to discuss their clinical and patho- logical significance in urothelial carcinoma of the bladder. Methods The expression of S6K1 and 4EBP-1 were de- tected in 80 urothelial carcinoma of the bladder tissues and 20 normal bladder mucosa tissues by Western blot. Re- sults The expression rate of S6K1 in urothelial carcinoma of the bladder tissues （64%） was significantly higher than that in normal bladder mucosa tissues （10V0）. The expression rate of 4EBP-1 in normal bladder mucosa tis- sues （15 ~） was significantly lower than that in urothelial carcinoma of the bladder tissues （71~）. The expres- sion rates of S6K1 and 4EBP-1 protein in low-grade bladder cancer were significantly higher than those in the high- grade bladder cancer （P〈0.05）. The expression rates of S6K1 and 4EBP-1 protein in recurrent tumors were high- er than those in the primary tumor （P〈0.05）. There was no significant difference of the expression rate of S6K1 and 4EBP-1 protein in patients with different gender, different tumor stage and with or without smoking （P〉0. 05）. Conclusion S6K1 and 4EBP-1 are closely related to the pathological grade and recurrence of urothelial carci- noma of the bladder, which may become new targets for cancer therapy.
